RNA-Seq analysis was carried out to investigate the effects of expression of wild-type CDK8 (CDK8) or CDK19 (CDK19) or their kinase-inactive D173A mutants (CDK8M and CDK19M) in HEK293 cells (WT) and their CDK8/19 double-knockout (dKO) derivatives
Overall design: Two different sets of the re-expression derivatives of dKO cells generated with pHIV-dTomato or pHIV-luc-Blast based lentiviral constructs (dKO-V, dKO-8, dKO-8M, dKO-19 and dKO-19M; dKO-V_2, dKO-8_2, dKO-8M_2, dKO-19_2 and dKO-19M_2) and a set of the over-expression derivatives of WT cells generated with pHIV-luc-Blast based lentiviral constructs (WT-V, WT-8, WT-8M, WT-8M, WT-19 and WT-19M) were analyzed by RNA-Seq for transcriptomic profiles under regular culture condition. Each condition (expression derivative) was done in biological triplicate or more.
