Guselkumab reduces disease- and mechanism-related biomarkers more than adalimumab in patients with psoriasis: a VOYAGE 1 substudy

(Submitter supplied) In the phase 3 VOYAGE-1 trial (ClinicalTrials.gov identifier: NCT02207231), guselkumab demonstrated superior efficacy versus placebo and the tumor necrosis factor (TNF)-α antagonist, adalimumab, in patients with moderate-to-severe plaque psoriasis (Blauvelt et al., 2017). Here, skin samples were collected from patients in VOYAGE-1 and pharmacodynamic (PD) responses to guselkumab (vs adalimumab) treatment were assessed over 48 weeks.

Micro RNA Expression data from DU145 shDCUN1D1

(Submitter supplied) Defective in Cullin Neddylation 1 Domain Containing 1 (DCUN1D1) is an E3 ligase for neddylation, a post translational process similar to, and that runs in parallel with the ubiquitin proteasome pathway. Although being established as an oncogene in various squamous cell carcinomas, the role of DCUN1D1 in prostate cancer has not been explored. Here we used microarray analysis to determine the set of genes regulated by DCUN1D1 in prostate cancer cells. more...

Transcriptional profiling of immunity to the MF-59 – adjuvanted influenza vaccine in children

(Submitter supplied) In this study, we performed a systems-level analysis of immune responses to the trivalent inactivated influenza vaccine adjuvanted with MF-59 in children (15-24 months old) and in young healthy adults. We analyzed transcriptional responses elicited by vaccination in peripheral blood, as well as cellular and antibody responses following primary and booster vaccinations. The pediatric population is a major target of vaccination, yet there is a paucity of studies on the transcriptional response of immunity to vaccination in this special population. more...

Defining molecular and cellular landscape behind non-response to Lupus Nephritis drugs

(Submitter supplied) Lupus nephritis (LN) is one of the more severe systemic lupus erythematosus manifestations with the potential of developing into end stage kidney disease. Mycophenolate mofetil (MMF) and Azathioprine (AZA) are widely used for both induction and maintenance therapy for LN, but the one year complete renal response ranges from 30-40% in most trials. Reasons for non-response are still unknown. Thus, anticipating lack of drug efficacy in a patient could lead to early introduction of advanced therapies. more...

In uveal melanoma Gα-protein GNA11 mutations convey a shorter disease-specific survival and are more strongly associated with loss of BAP1 and chromosomal alterations than Gα-protein GNAQ mutations

(Submitter supplied) Mutations in the Gα-genes GNAQ and GNA11 are found in 85-90% of uveal melanomas (UMs). We analyzed the association between GNAQ and GNA11 mutations with disease-specific survival, gene expression profiles, and cytogenetic alterations in 219 UMs. Our analysis showed a shorter disease-specific survival of GNA11-mutated cases as compared to those carrying a GNAQ mutation (HR=1.97 [95%CI 1.12-3.46], p=0.02). more...

ASPSCR1-TFE3 orchestrates the angiogenic program of alveolar soft part sarcoma I

(Submitter supplied) Alveolar soft part sarcoma (ASPS) is a rare soft part malignancy affecting adolescents and young adult. ASPS is characterized by its alveolar structure consisting of tumor cells and highly integrated vascular network, and its high metastatic potential indicates the importance of the prominent angiogenic activity of ASPS. Here we find that the expression of ASPSCR1-TFE3, the fusion transcription factor causatively associated with ASPS, is dispensable for in vitro tumor maintenance but required for in vivo tumor development via angiogenesis. more...

Cerivastatin synergizes with trametinib and enhances its efficacy in the therapy of Uveal Melanoma

(Submitter supplied) Background and Aims: Uveal melanoma (UM), the most common primary intraocular tumor in adults, is characterized by marked variability in its ability to metastasize. In fact, up to half the patients with UM develop distant metastases, most commonly to the liver. Mutations in alpha G-protein subunits, GNAQ and GNA11, are found in most primary UM, in a mutually exclusive pattern. Also, mutations in SF3B1, EIF1AX, and BAP1, are associated with markedly distinct prognoses Targeted therapy with MAP kinase inhibitors has been unsuccessful in the treatment of uveal melanoma (UM). more...

Platinum-sensitive HGOC residual tumors share joint expression of CRYAB, CEACAM6, SOX2 genes with tumors that acquired resistance

(Submitter supplied) Purpose: Most High-Grade Ovarian Carcinomas (HGOCs) are sensitive to carboplatin (CBP)-based chemotherapy but frequently recur within 24 months. Recurrent tumors remain frequently CBP-sensitive and acquire resistance after several CBP cycles. We developed Patient-Derived Xenografts (PDXs) that allow the study of these different stages of CBP-sensitive recurrence and acquisition of resistance. Experimental Design: We generated PDX models from CBP-sensitive HGOC. more...

Systems Biology of Influenza A (H5N1) Virus Monovalent Vaccine with and without AS03 adjuvant

(Submitter supplied) H5N1 influenza vaccine is known to induce diminished antibody responses in vaccine recipients without the use of adjuvant. We hypothesize that this is due to intrinsic defects in innate responses that could explain the poor immunogenicity of the vaccine. These innate responses may be improved with use of an adjuvant, e.g., AS03.

Distinct peripheral blood molecular signature emerges with successful tacrolimus withdraw in kidney transplant recipients

(Submitter supplied) Tacrolimus (Tac) is an effective anti-rejection agent in kidney transplantation, but its off-target effects make withdrawal desirable. While studies indicate that Tac can be safely withdrawn in a subset of kidney transplant recipients, immune mechanisms that underlie successful vs. unsuccessful Tac removal are unknown. We performed microarray analyses of PBMC RNA from subjects enrolled in the Clinical Trials in Organ Transplantation-09 study in which stable kidney transplant recipients were randomized to Tac withdrawal or maintenance of standard immunosuppression beginning 6-mo post-transplant. more...

Transcriptome analysis defines ToF and ASD myocardial gene signatures and reveals disease-specific gene reprogramming upon surgery with cardiopulmonary bypass

(Submitter supplied) Tetralogy of Fallot (ToF) and Atrial Septal Defects (ASD) are the most common types of congenital heart disease and a major cause of childhood morbidity and mortality. Cardiopulmonary bypass (CPB) is used during corrective cardiac surgery to support circulation and heart stabilization. However, this technique triggers systemic inflammatory and stress response and consequent increased risk of postoperative complications. more...
