LSK immunophenotype hematopoietic stem cells after irradiation at day points in 3 weeks

(Submitter supplied) In this study, we aimed to investigate the differentiation dynamics of LSK (Lin-Sca1+c-Kit+) cells following radiation injury. LSK cells, which are a critical component of the hematopoietic stem and progenitor cell population, play a vital role in the regeneration and repair of the hematopoietic system after damage. By utilizing these cells, we sought to understand the mechanisms underlying their response to radiation, focusing on their capacity for self-renewal and differentiation into various blood cell lineages.

Low input RNA Sequencing of effector and naive WT and NCOR1-cKO CD4+ Treg cells

(Submitter supplied) Analysis of Treg cells isolated from WT and NCOR1-cKO mice indicated an important function of NCOR1 in regulating the transition from naïve to effector Treg cells. The aim of this NGS experiment was to determine the transcriptome profile of ex vivo isolated naïve and effector WT and NCOR1-cKO Treg cells under steady state condition.

Targeting HDAC1 enzymatic activity in T cells protects against experimental autoimmune encephalomyelitis

(Submitter supplied) Histone deacetylases are key epigenetic regulators that control T cell-mediated immunity. A T cell-specific deletion of Hdac1 (HDAC1cKO) protects mice against experimental autoimmune encephalomyelitis (EAE). However, it remains elusive whether inhibition of HDAC1 enzymatic activity and thus mimicking HDAC1 inhibitor treatment is sufficient to block EAE induction. In order to address this question, we generated a novel mouse strain that expresses catalytically inactive HDAC1 (HDAC1Off) from the Rosa26 locus in HDAC1cKO CD4+ T cells to mimic selective inhibition of HDAC1 enzymatic activity in vivo. more...

Lack of TYK2 Signaling Enhances Host Resistance to Candida albicans Skin Infection

(Submitter supplied) Candida albicans is the most common human fungal pathogen, causing diseases ranging from local to life-threating systemic infections. Tyrosine kinase 2 (TYK2), a crucial mediator in several cytokine signaling pathways, has been associated with protective functions in various microbial infections. However, its specific contribution in the immune response to fungal infections has remained elusive. In this study, we show that mice lacking TYK2 or its enzymatic activity exhibit enhanced resistance to C. more...

LSK immunophenotype hematopoietic stem cells after irradiation at day 2

(Submitter supplied) To investigate the impact of radiation damage on hematopoietic stem cell fate decision, we conducted a longitudinal study, collecting single-cell data on days 2 post-radiation exposure. This approach enabled us to observe changes in the subpopulations of hematopoietic stem cells at the LSK level in mice over time. Additionally, we assessed alterations in their fate determination and lineage differentiation tendencies throughout the recovery process.

Remodeling of Il4-Il13-Il5 locus underlies selective gene expression (ChIP-seq)

(Submitter supplied) The type 2 cytokines, interleukin (IL)-4, IL-5 and IL-13 reside within a multi-gene cluster in mammals. These cytokines represent the hallmark of type 2 immune responses controlling parasites and promoting tissue repair, as well as causing allergic diseases. Both innate and adaptive lymphocytes secrete type 2 cytokines with discordant production spectra. We took a holistic structural and functional view of the type 2 cytokine locus before and after activation, comparing innate (ILC2) and adaptive (Th2) lymphocytes to understand mechanisms underlying their distinctive programs. more...

(Submitter supplied) The goals of this study are to compare gene expression profiles of Setd2 WT and KO germinal center B cells and reveal the underlying mechanisms by which Setd2 regulates germinal center B cell responses. Further our study aims to compare gene expression profiles of Phf19 WT and KO germinal center B cells reveal the underlying mechanisms by which Phf19 regulates germinal center B cells responses. Finally, we combine the two profiles and investigate the relation shape betweeen Setd2 dependent regulation and Phf19 dependent regulation.

Interaction between subventricular zone microglia and neural stem cells impacts the neurogenic response in a mouse model of cortical ischemic stroke

(Submitter supplied) After a stroke, the neurogenic response from the subventricular zone (SVZ) to repair the brain is limited. Microglia, as an integral part of the distinctive SVZ microenvironment, control neural stem / precursor cell (NSPC) behavior. Here, we show that discrete stroke-associated SVZ microglial clusters negatively impact the innate neurogenic response, and we propose a repository of relevant microglia–NSPC ligand–receptor pairs. more...

Single Cell RNA sequencing of CD11b+Gr1+ cells from the bone marrow of young and aged mice

(Submitter supplied) Myeloid-derived suppressor cells (MDSCs) are recognized as one of the key mediators of immunosuppression in aging process, which induce immunosenescence and increase elderly people’s susceptibility to to infections, cancers, autoimmune diseases, and degenerative diseases. It still remains elusive whether MDSCs represent a unique subset of myeloid cells that differ from their young, healthy counterparts, which will hamper the deeper understanding of the pathophysiological functions of MDSCs. more...

Characterization of ribosome stalling and no-go mRNA decay stimulated by the Fragile X protein, FMRP

(Submitter supplied) Loss of functional fragile X mental retardation protein (FMRP) causes fragile X syndrome (FXS) and is the leading monogenic cause of autism spectrum disorders and intellectual disability. FMRP is most notably a translational repressor and is thought to inhibit translation elongation by stalling ribosomes as FMRP-bound polyribosomes from brain tissue are resistant to puromycin and nuclease treatment. more...

Tff1-expressing Tregs in lung prevents exacerbation of Bleomycin-induced pulmonary fibrosis (scRNA-Seq)

(Submitter supplied) Bleomycin (BLM) induces lung injury, leading to inflammation and pulmonary fibrosis. Regulatory T cells (Tregs) maintain self-tolerance and control host immune responses. However, little is known about their involvement in the pathology of pulmonary fibrosis. Here we show that a unique Treg subset that expresses trefoil factor family 1(Tff1) emerges in the BLM-injured lung. These Tff1-expressing Tregs (Tff1-Tregs) were induced by IL-33. more...
