Gene expression dynamics reveal Sprouty mediated regulation of Wnt pathway genes

(Submitter supplied) Sprouty genes (Spry) are feedback regulators of receptor tyrosine kinase signaling, and are genes with known tumor suppressing activity. The impact of Spry induction is ligand specific, but it is well-understood to be a negative feedback regulator of the mitogen-activated protein kinase pathway in response to fibroblast growth factor (FGF). However, the precise impact Spry has on transcription resulting in tumor suppression remains unknown. more...

Microarray expression analysis depicting MHCII-dependent features of FOXP3-expressing CD4 T regulatory cells

(Submitter supplied) Analysis comparing K14 and MCHII+/- Tregs. The hypothesis was that the lack of MHCII+/- in the spleen and intestine would result in a large shift in K14 Treg gene expression away from an activated and effector phenotype. Results demonstrate how a lack of MHCII-antigen presentation in the small intestine, where Tregs exhibit an effector penotype, does not greatly reduce effector features of T regulatory cells in K14 mice.

Defining the Sox2-independent reprogramming steps to pluripotency

(Submitter supplied) Several Sox family members, small molecules, and inhibitors of the TGF-β pathway can replace exogenous Sox2 during the generation of induced pluripotent stem cells (iPSCs), suggesting that SOX2 is dispensable for initiation of reprogramming. However, the time point at which the endogenous Sox2 locus is activated in these conditions was not thoroughly evaluated. To identify the Sox2-independent reprogramming steps, we excluded Sox2 from the reprogramming cocktail and used Sox2-deficient mouse embryonic fibroblasts. more...

Prospective identification and characterization of multipotent peripheral neural stem cells out of mammalian central nervous system

(Submitter supplied) It has been widely accepted that mammalian neural stem cells (NSCs) only exist in central nervous system (CNS). Here, we challenge this concept and show that peripheral NSCs (pNSCs) could be isolated out of CNS from mouse embryonic limbs, postnatal tail and adult lung tissues. Derived-pNSCs express multiple NSC-specific markers, exhibit cell morphology, self-renewing capacity, genome-wide transcriptional profile, epigenetic features similar to those of brain control NSCs. more...

The transcriptional signature for Hepatocyte Growth Factor-driven invasive growth predicts poor prognosis of human hepatocellular carcinoma.

(Submitter supplied) During embryogenesis, Hepatocyte Growth Factor (HGF) elicits a distinctive morphogenetic program, the invasive growth, by the activation of MET, whose aberrant activation in cancer drives metastatic progression. Aim of this work is to define and characterize the transcriptional signature of invasive growth, and to verify its activation in human cancers. Global expression profiling was carried out on mouse liver stem/progenitor cells (MLP-29) stimulated for different times, one, six and twenty-four hours, in vitro with HGF to define the invasive growth signature. more...

Tissue resident memory T cells trigger rapid exudation and local antibody accumulation

(Submitter supplied) Adaptive immunity is didactically partitioned into humoral and cell-mediated effector mechanisms, which may imply that each arm is separate and does not function together. Here, we report that the activation of CD8+ resident memory T cells in nonlymphoid tissues triggers vascular permeability, which facilitates rapid distribution of serum antibodies into local tissues. TRM reactivation was associated with transcriptional upregulation of antiviral signaling pathways as well as Fc receptors and components of the complement cascade. more...

Gene expression profiling of MYC inducible murine tumor models (tissues) [microarray]

(Submitter supplied) The transcription factor, c-MYC, is a proto-oncogene that is activated in many types of cancer. To study how MYC promotes and maintains the neoplastic phenotype, we use tetracycline-regulated systems in transgenic mice whereby we can conditionally activate MYC. We profile the changes in gene expression when MYC is activated versus inactivated in order to identify a gene signature that is associated with the ability of MYC to promote tumorigenesis.

Gene expression profiling of Calca neurons in the parabrachial nucleus (PBN)

(Submitter supplied) Calca-expressing neurons in the parabrachial nucleus (PBN) play a major role in regulating appetite and transmitting real or potential threat signals. Here, we reveal transcripts specific or highly enriched in this neuronal population by using mouse genetics and a conditional viral ribosome-tagging approach to phenotype these neurons.

Comparative study of transcriptome in the hearts isolated from mice, rats, and human

(Submitter supplied) The heart is a major organ in mammalian life, and the study of transcription factors, particularly those that regulate cardiac rhythm, has been an important focus of science. However, few studies have focused on species differences. Accordingly, challenges remain in studying genes that have universal functions across species and genes that determine species differences. In this study, we reanalyzed mouse, rat, and human transcriptome data obtained from different platforms by calculating specificity measure (SPM) values in consideration of species differences. more...

mRNA expression profile of regulatory neutrophil induced by sodiumtaurodeoxycholate

(Submitter supplied) Conventional strategies for the development of new drugs have used chemical compounds or large molecules that act as an agonist, antagonist, inhibitor (enzyme or signal transducer), enzyme cofactor or supplement for normal constituents in the body. Recently, the field of cell therapeutics has received much attention. Ex vivo expansion and/or manipulation is essential in cell therapy, which might limit the entrance of pharmaceutical companies into the cell therapy market. more...

Doublecortin mutation leads to persistent defects in the Golgi apparatus and mitochondria in adult hippocampal pyramidal cells

(Submitter supplied) Human doublecortin (DCX) mutations are associated with severe brain malformations leading to aberrant neuron positioning (heterotopia), intellectual disability and epilepsy. DCX is a microtubule-associated protein which plays a key role during neurodevelopment in neuronal migration and differentiation. Dcx knockout (KO) mice show disorganized hippocampal pyramidal neurons. The CA2/CA3 pyramidal cell layer is present as two abnormal layers and disorganized CA3 KO pyramidal neurons are also more excitable than wild-type (WT) cells. more...

Relative contributions of sex hormones, sex chromosomes, and gonads to sex differences in tissue gene regulation

(Submitter supplied) We systemetically distinguished the effects of each class of sex-biasing factors on transcriptome regulation in liver and adipose tissues with the Four Core Genotype (FCG) mouse model. The results offers a tissue-specific account of the individual contribution of major sex-biasing factors to gene regulation.

A balanced Oct4 protein interaction network is crucial for maintaining pluripotency

(Submitter supplied) Oct4 is considered a master transcription factor for pluripotent cell self-renewal and embryo development. It primarily collaborates with other transcriptional factors or coregulators to maintain pluripotency. However, it is still unclear how Oct4 interacts with its partners. Here, we show that the Oct4 linker interface mediates competing and balanced Oct4 protein interactions which are crucial for maintaining pluripotency. more...

Gene expression microarray analysis of whole mouse hamstring muscles to identify genes promoting the devlopment of neurogenic heterotopic ossifications following spinal cord injury

(Submitter supplied) To better understand which genes/pathways are activated in injured muscles following a spinal cord injury (SCI) and how these promote the development of neurogenic heterotopic ossification (NHO), we performed gene expression profiling by microarray of muscles in the early phase following muscle injury.
