Origin of replication for Pyrococcus abyssi by ChIP-chip

(Submitter supplied) Chromatin immunopreciptation coupled with hybridization on a whole genome microarray (ChIP-chip) of P. abyssi. Protocol: Microarray preparation. The P. abyssi genome was divided into 353 regions, each of which is 5 kb in length. Each of the 353 DNA regions has a specific ID number. For example, the microarray region 25 corresponds to the genome coordinates 120 000 to 125 000. For the microarrays, each of the 353 regions was amplified with the BD Advantage 2 PCR kit (BD biosciences clontech). more...

MH Hannover Pseudomonas putida 4.5K

(Submitter supplied) Purified DNA of some 4600 overlapping shotgun clones in vector pCR-BluntII-TOPO (Invitrogen, Groningen, The Netherlands) was used as template in standard PCR amplification with the vector-specific primer pair d(TCGGATC CACTAGTAACG) and d(GGCCGCCAGTGTGATG). Shotgun cloning had been performed according to the procedures described by the vector's manufacturer. Each PCR reaction was individually checked by agarose gel electrophoresis. more...

GenoToul/Biopuces operon_yeast22x22

(Submitter supplied) Dendrislides are prepared according to Trevisiol et al. 2003, New J. Chem. 27, 1713-1719 Protocol: Oligos sold by Operon 70-mer NH2 modified, put in phosphate buffer 0.3 M, pH 9 at 20 µM spotted on dendrislide (Le Berre et al. Nucleic Acids Research, 2003, Vol. 31, No. 16 e88). Robot is ChipWriterPro (Biorad) with 4x8 pins each pin has spotted a block of 22x22 spots. Each well is spotted in duplicate.

Gene-environment interaction in yeast gene expression

(Submitter supplied) This study is a quantitative trait linkage mapping study targeted at gene-environment interaction. A family of yeast derived from BY4716 and RM11-1a was grown in two carbon sources (glucose and ethanol) and expression profiled. The analysis revealed how the influence of genetic factors changes in different conditions. Abstract: The effects of genetic variants on phenotypic traits often depend on environmental and physiological conditions, but such gene–environment interactions are poorly understood. more...
