AML1-ETO fusion protein effect on CD34+ hematopoietic cells

Analysis of CD34+ hematopoietic cells expressing the AML1-ETO fusion protein which is formed in acute myeloid leukemia (AML) from translocation t(8;21). CD34+ cells isolated from peripheral blood progenitor and cord blood cells. Results provide insight into the role of AML1-ETO in leukemogenesis.

AML1-ETO-induced FOXO1 activates a self-renewal program in pre-leukemia stem cells

(Submitter supplied) Blocking the self-renewal of pre-leukemia stem cells could prevent AML relapse. In this study we show that FOXO1 is an essential self-renewal factor in leukemic and pre-leukemic cells expressing the t(8;21)-associated oncogene AML1-ETO (AE). FOXO1 is consistently upregulated in t(8;21) AML and functions as a critical oncogenic mediator rather than a tumor suppressor. Expression of FOXO1 in human CD34+ cells promotes a pre-leukemic state, partially phenocopying the cellular and transcriptional effects of AE expression. more...

AML1/ETO accelerates cell migration and impairs cell-to-cell adhesion and homing of hematopoietic stem/progenitor cells

(Submitter supplied) The AML1/ETO fusion protein found in acute myeloid leukemias functions as a transcriptional regulator by recruiting co-repressor complexes to its DNA binding site. In order to extend the understanding of its role in preleukemia, we expressed AML1/ETO in a murine immortalized pluripotent hematopoietic stem/progenitor cell line, EML C1, and found that genes involved in functions such as cell-to-cell adhesion and cell motility were among the most significantly regulated as determined by RNA sequencing. more...

In Vitro Transformation of Primary Human CD34+ Cells by AML Fusion Oncogenes: Early Gene Expression Profiling Reveals Possible Drug Target in AML

(Submitter supplied) Different fusion oncogenes in acute myeloid leukemia (AML) have distinct clinical and laboratory features suggesting different modes of malignant transformation. Here we compare the in vitro effects of representatives of major groups of AML fusion oncogenes on primary human CD34+ cells.

Acute myeloid leukemia fusion oncogenes in vitro effect on primary CD34+ cells: time course

Analysis of CD34+ cells nucleofected (6h) or transduced (3d, 8d) with AML leukemia fusion oncogenes: AML1-ETO, MLL-AF9, PML-RARA or NUP98-HOXA9. Results provide insight into molecular mechanisms of fusion oncogene-driven leukemogenesis and early temporal patterns of gene deregulation in AML.

Gene expression signature of MYB-GATA overexpression plus GATA1 down regulation in undifferentiated or differentiated UT7 cells

(Submitter supplied) Acute basophilic leukemia is a rare subtype of acute myeloblastic leukemia. We recently described a recurrent t(X;6)(p11;q23) translocation occurring in male infants with acute basophilic leukemia, generating a MYB-GATA1 fusion gene. To better understand the role of MYB-GATA1 in the leukemogenesis of acute basophilic leukemias, we expressed this chimeric transcription factor in the UT-7 cell line under normal or differentiating conditions. more...
