Neurogenic potential of mesenchymal stem cells

Comparison of mesenchymal stem cells (MSC) treated with dimethylsulphoxide and butylated hydroxyanisole (DMSO/BHA) for 6 or 48 hours to fetal brain (positive control) or adult liver (negative control). Results raise doubts about the DMSO/BHA protocol for generating neural cells from MSCs.

Neurogenic potential of human mesenchymal stem cells revisited

(Submitter supplied) The possibility of generating neural cells from human bone marrow-derived mesenchymal stem cells by simple in vitro treatments is alluring both conceptually and practically. However, whether phenotypic modulations observed after chemical manipulation of such stem cells truly represent a genuine trans-lineage differentiation remains to be established. We have re-evaluated the effects of a frequently reported biochemical approach, based on treatment with butylated hydroxyanisole and dimethylsulphoxide, to bring about such phenotypic conversion by monitoring the morphological changes induced by the treatment in real time, by analysing the expression of phenotype-specific protein markers and by assessing the modulation of the cells’ transcriptome. more...

Micro-CRIBI Human Oligo Array (Operon V2.0)

(Submitter supplied) This set consists of 21,329 70mer oligonucleotides (Operon version 2.0) designed on Human Unigene clusters, mainly in the 3 end terminal region. Each oligonucleotide is spotted by our robotic station Biorobotics Microgrid II, in two replicates on a MICROMAX glass slide SuperChip I (Cat No MPS696) provided by PerkinElmer Life Sciences Inc. The deposition is qualitatively assessed by a scan of the spotted slide at 10 micron resolution, using a GSI Lumonics scanner and a protocol that allows the detection of spotted features through the salts derived from the dried solution in which oligonucleotides are dissolved. more...

Meta analysis of gene expression in human islets after in vitro expansion.

(Submitter supplied) Pancreatic islet transplantation as a cure for type 1 diabetes (T1D) cannot be scaled up due to a scarcity of human pancreas donors. In vitro expansion of beta cells from mature human pancreatic islets provides an alternative source of insulin-producing cells. The exact nature of the expanded cells produced by diverse expansion protocols, and their potential for differentiation into functional beta cells, remain elusive. more...

In vitro expanded and re-differentiated pancreatic islet cells

Analysis of fresh pancreatic islets, expanded islet cells, and re-differentiated cells. Three different protocols were used to expand pancreatic islets, followed by attempts at redifferentiation back to islets. Results provide insight into active pathways in expanded and redifferentiated islets.

Overview of gene expression alternatively modulated during the differentiation of human embryonic stem cells (hES)

(Submitter supplied) The transcriptome analysis was performed in triplicate using two human embryonic stem cells lines (hES_VUB01 and hES_SA01) by comparing the expression profiles of the undifferentiated hES cells and two types of progenitors derived from the hES cell lines: Neural progenitors (NPC) and Mesodermal progenitors (MSC). Comparative transcriptome analysis of dedifferentiation in porcine mature adipocytes and follicular granulosa cells

(Submitter supplied) Cellular dedifferentiation signifies the withdrawal of cells from a specific differentiated state into a ‘stem cell’-like undifferentiated state. However, the mechanism of dedifferentiation remains obscure. We showed that mature adipocytes (MA) and follicular granulosa cells (GC), which have distinct functions in vivo, can dedifferentiate during culture in vitro and acquire multipotency. We investigated the dedifferentiation mechanism of MA and GC using global gene expression analyses.

Comparison study of glioma stem cells and adult human neural stem cells

(Submitter supplied) Glioblastoma, the most common and malignant brain tumor, harbors stem-like cells that self-renew and propagate upon serial transplantation. Although they share functional, morphological and developmental similarities to adult brain neural stem cells, stem cell characteristic pathways contributing to glioblastoma stem-like cells have not been consistently determined. Towards this goal we have provided an internally coherent molecular reference that compares adult neural and glioblastoma stem cells cultivated under identical conditions. more...
