Transcriptome of a mouse kidney cortical collecting duct cell line: effects of aldosterone and vasopressin

(Submitter supplied) In this study, we analyzed the transcriptome of a highly differentiated mouse clonal CCD principal cell line (mpkCCD(cl4)) and the changes in the transcriptome induced by aldosterone and vasopressin. Serial analysis of gene expression (SAGE) was performed on untreated cells and on cells treated with either aldosterone or vasopressin for 4 h. The transcriptomes in these three experimental conditions were determined by sequencing 169,721 transcript tags from the corresponding SAGE libraries. more...

Effects of water restriction on gene expression in mouse renal medulla

(Submitter supplied) In order to identify novel gene targets of vasopressin regulation in the renal medulla, we performed a cDNA microarray study on the inner medullary tissue of mice following a 48 hour water restriction protocol. Keywords: Stress Response

Expression data from mouse collecting duct cell, mpkCCD, in response to a peptide hormone vasopressin analog, dDAVP

(Submitter supplied) Vasopressin is the major hormone that regulates renal water excretion. It does so by binding to a receptor in renal collecting duct cells, triggering signaling pathways that ultimately regulate the abundance, location, and activity of the water channel protein aquaporin 2. We took an advantage of quantitative large scale proteomic technologies and oligonucleotide microarrays to quantify steady state changes in protein and transcript abundances in response to vasopressin in a collecting duct cell line, mpkCCD clone 11 (Yu et al. more...

Aldosterone-responsive genes in kidney cortical collecting duct cell line, mpkCCDc14

(Submitter supplied) Aldosterone is arguably the single most important hormone implicated in the control of blood pressure and extracellular fluid volume in mammals. It acts primarily by increasing the rate of transepithelial sodium transport in the kidney tubules. Several monogenetic defects resulting in hypertension have now been attributed to abnormalities in sodium handling within the aldosterone-sensitive distal nephron, where the epithelial sodium channel, ENaC, constitutes the rate-limiting step of sodium transport. more...

Qiagen/Operon version 2.0 mouse oligonucleotide

(Submitter supplied) The Qiagen/Operon version 2.0 mouse oligonucleotide library, comprised of oligonucleotides (70-mers) representing 16,443 unique mouse genes, was printed on poly-L-lysine-coated glass slides. The library also included sets of positive and negative controls that were used for quality control purposes. Oligonucleotides were prepared in sets of 384-well plates at a concentration of 40 <E6>M. Arrays were prepared using a custom-built array printer equipped with Majer MicroQuill-2000 split tip pins with a capacity of 255 slides per print run. more...

Aldosterone effect on kidney cortical collecting cell line: time course

Analysis of kidney epithelial cortical collecting duct cells treated with aldosterone for 1 or 6 hours. Aldosterone stimulates Na+ transport in tight epithelia. Results identify candidate genes that regulate Na+ transport via the epithelial Na+ channel and mediate the early response to aldosterone.

Murine inner medullary collecting duct cells response to aldosterone

(Submitter supplied) Murine inner medullary collecting duct cells were treated for 1 hour with vehicle (control) or aldosterone. Total RNA was isolated and used as template to generate the eventual cRNA target. The experiment was repeated a total of three times. Six cRNA samples, three control and three treated, were generated and used in a total of six hybridizations. The mineralocorticoid aldosterone is a major regulator of Na+ and acid-base balance and control of blood pressure. more...
