Bleomycin exposed lymphoblastoid cells

(Submitter supplied) The uncovering of genes involved in susceptibility to the sporadic cancer types is a great challenge. It is well established that the way in which an individual deals with DNA damage is related to the chance to develop cancer. Mutagen sensitivity is a phenotype that reflects an individual’s susceptibility to the major sporadic cancer types, including colon, lung and head and neck cancer. A standard test for mutagen sensitivity is measuring the number of chromatid breaks in lymphocytes after exposure to bleomycin. more...

Bleomycin effect on mutagen-sensitive lymphoblastoid cell lines

Analysis of mutagen-sensitive lymphoblastoid cell lines after exposure to bleomycin. Mutagen-sensitive cells exhibit a high number of bleomycin-induced chromatid breaks. Mutagen sensitivity (MS) reflects an individual's susceptibility to sporadic cancers. Results identify genes involved in MS.

Survival Prediction of HNSCC using FFPE samples

(Submitter supplied) Recently it was shown that gene expression signatures generated from DNA microarray analyses have promise as biomarkers of clinical outcome and that the molecular characteristics of tumors could be elucidated. Through this study, we have determined a high-risk signature for recurrence as a prognostic biomarker using formalin-fixed HNSCC tumors and tested the results to an independent data set obtained from fresh frozen tumors as a comparison. more...

Gene expression profiling reveals novel molecular marker candidates of laryngeal squamous cell carcinoma

(Submitter supplied) Laryngeal squamous cell carcinoma is a very common in head and neck cancer, accounting for 25% of all cases, with high mortality rates and poor prognosis. In this study, we compared expression profiles of clinical samples from15 larynx tumors and 10 non-neoplastic larynx tissue using a custom-built cDNA microarray containing 332 probes for 285 genes previously identified as up- or down-regulated in head and neck tumors by the Head and Neck Annotation Consortium (Reis et al., Cancer Res 65:1693-99, 2005). more...

Chromosomal aberration of ESCC and HSCC cell lines

(Submitter supplied) In order to identify putative targets in squamous cell carcinoma (SCC), a survey of parallel chromosomal alterations and gene expression studies in 10 SCC cell lines were performed using array-CGH and microarray techniques. The most frequent changes were gains of 11q13.1-13.3 and losses of 18q12.1-23 in SCC. Furthermore, the expression levels of the sets of genes at both these loci in SCC were measured using microarray analysis. more...

Mat-Lylu cell line compared to G cell line

(Submitter supplied) we analyzed the gene expression profiles of Mat-Lylu cell lines (in duplicate) compared to G cell lines (in duplicate) using Affymetrix tools and dChip software. The objective was to find metastasis-associated genes in prostate cancer, using this in vitro model. Keywords: cell line comparison

Expression Changes Relevant for Apoptosis in K562 Cells Co-Treated with Amifostine and Imatinib

(Submitter supplied) We had previously demonstrated an increased proapoptotic effect of Imatinib (STI571) in combination with Amifostine (AMI) in K562 cell line. In this study we used genomic scale gene expression profiling to monitor changes at transcriptional level in K562 cells during the treatment with AMI+STI571. We identified a transcriptional repressor of survival genes, known as BTF, which triggers a pro-apoptotic signal, potentially helpful to overcome the resistance to STI571. more...

Comparative study of gene expression in human colorectal cancer tissues and normal mucosa

(Submitter supplied) The purpose of our study was to better understand the genetic mechanism of oncogenesis for human colorectal cancer and to identify potential new tumour markers of use in clinical practice. Results: Comparing cancerous tissues with normal colonic mucosa we identified 584 known genes (3%) differentially expressed to a significant degree (p<0.001). Many of the transcripts that were more abundant in tumours than in non-neoplastic tissues appear to reflect important events for colon carcinogenesis. more...

Experimental condition

(Submitter supplied) BY4730 cultures were exposed to medium containing dilutions of test agent (Cisplatin (Cis), 600mg/mL; Methylmethane sulfonate (MMS), 0.12% v/v; Bleomycin (Bleo), 0.15U/mL; NaCl, 10mg/mL; ethanol, 15% v/v) for 120 min. Control cultures were either mock-treated with the solvent, DMSO (for comparison to Cis, MMS, Bleo) or maintained in media (for comparison to ethanol and NaCl) for 120 mins. Sample preparation and processing procedures were performed according the Affymetrix Genechipâ Expression Analysis Technical Manual (Affymetrix, Santa Clara, CA). more...

Identifying pH independent hypoxia induced genes in human squamous cell carcinomas in vitro

(Submitter supplied) Background: Genes upregulated by low oxygen have been suggested as endogenous markers for tumor hypoxia. Yet, most of the genes investigated have shown inconsistent results, which have led to concerns about their ability to be true hypoxia markers. Previous studies have demonstrated that expression of hypoxia induced genes can be affected by extracellular pH (pH e ). Methods: Five different human cell lines (SiHa, FaDu DD, UTSCC5, UTSCC14 and UTSCC15) were exposed to different oxygen concentrations and pH (7.5 or 6.3), and gene expression analyzed with microarray (Affymetrix - Human Genome U133 Plus 2.0 Array). more...
