Genome-wide transcriptional profiling of Lactobacillus acidophilus' response to bile

(Submitter supplied) Lactobacillus acidophilus NCFM is an industrially important strain used extensively as a probiotic culture. Tolerance of the presence of bile is an attribute important to microbial survival in the intestinal tract. A whole-genome microarray was employed to examine the effects of bile on the global transcriptional profile of this strain, with the intention of elucidating genes contributing to bile tolerance. more...

Temporal gene expression and probiotic attributes of Lactobacillus acidophilus during growth in milk

(Submitter supplied) Lactic acid bacteria have been used as starter strains in the production of fermented dairy products for centuries. Lactobacillus acidophilus is a widely recognized probiotic bacteria commonly added to yogurt and used in dietary supplements. In this study, a whole genome microarray was employed to monitor gene expression of L. acidophilus cells propagated in 11% skim milk (SM) during early, mid and late logarithmic phase, and stationary phase. more...

Identification of candidate genes involved in fructooligosaccharides (FOS) utilization by Lactobacillus paracasei

(Submitter supplied) A shotgun microarray-based approach was used to identify candidate genes encoding the FOS utilization pathway in L. paracasei 1195. Differential expression profiles between cells grown on FOS and glucose provided a basis for identifying genes specifically induced by FOS. In addition, transcriptional analysis of cells grown on FOS or FOS + glucose allowed us to investigate the effect of catabolite repression on the expression of the FOS-induced genes. more...

Genome-wide transcriptional profiling of Lactobacillus acidophilus stress response

(Submitter supplied) We used a whole genome array containing 97.4 % of the annotated genes of Lactobacillus acidophilus NCFM, a probiotic culture that belongs to the lactic acid bacteria group, to identify genes that are differentially expressed under several stress conditions. The Bile Response Repressor, BreR, Regulates Expression of the Vibrio cholerae breAB Efflux System Operon

(Submitter supplied) Enteric pathogens have developed several resistance mechanisms to survive the antimicrobial action of bile. We investigated the transcriptional profile of Vibrio cholerae O1 El Tor strain C6706 under virulence gene inducing conditions, in the presence and absence of bile. Microarray analysis revealed that the expression of 119 genes was affected by bile. The mRNA levels of genes encoding proteins involved in transport was increased in the presence of bile, whereas mRNA levels of genes encoding proteins involved in pathogenesis and chemotaxis was decreased. more...

Comparative gene expression study of Lactobacillus acidophilus NCFM and upp mutant (NCK1909)

(Submitter supplied) A L. acidophilus NCFM mutant (NCK1909) carrying an in-frame deletion within the upp gene (LBA0770) was constructed as a background host for a newly developed upp-based counterselectable gene replacement system for L. acidophilus. Since the NCK1909 mutant serves both as the host for genetic exchange and the reference strain for subsequent phenotypic studies of deletion mutants that will be generated from the counterselective gene replacement system, comparative gene expression study was performed to ensure the genotype of NCK1909 is representative of the NCFM parent strain. more...

Lactobacillus reuteri ATCC 55730: Before vs after exposure to 0.5% bile.

(Submitter supplied) Transcriptional profiling of Lactobacillus reuteri ATCC 55730 mid-log cultures before vs after exposure to 0.5% bovine bile (oxgall). Two sets of array experiments were performed. One set compared the expression profiles of L. reuteri ATCC 55730 cells before bile exposure vs cells that had been exposed to 0.5% bile for 15 minutes (bile shock). The other set compared the expression profiles of L. reuteri ATCC 55730 cells before bile exposure vs cells that had begun growing again in the presence of 0.5% bile (bile adaptation). more...
