Differential gene expression during porcine conceptus trophoblastic elongation and attachment to the uterine epithelium

(Submitter supplied) The objective of the present investigation was to utilize the GeneChip® Porcine Genome Array from Affymetrix possessing 20, 201 unique probe sets to identify differentially expressed genes during rapid trophoblastic elongation and attachment to the uterine surface in the pig. Identification and characterization of conceptus gene expression patterns during rapid trophoblastic elongation and attachment in the pig will provide a better understanding of the events required for successful implantation and embryonic survival.

Pig peri-implantation embryo development transcriptional profiling

(Submitter supplied) A novel porcine cDNA microarray, containing PCR products selected for embryonic expression, was used for transcriptional profiling during conceptus development. Total RNA from quadruplicate samples of small spherical (1-5mm), large spherical (6-11mm), tubular (13-45mm), and filamentous (>100mm) was extracted, amplified, and converted to 32P labeled target and hybridized to membranes. Porcine peri-implantation embryo development order is from small spherical to large spherical, then to Tubular, then to filamentous. more...

Deep sequencing of the porcine endometrial transcriptome on day 12 of pregnancy

(Submitter supplied) The porcine conceptus undergoes rapid differentiation and expansion of its trophoblastic membranes between Days 11 and 12 of gestation. Concomitant with trophoblast elongation, production of conceptus estrogen, the porcine embryonic pregnancy recognition signal increases. Conceptus attachment to the uterine surface epithelium starts after Day 13 initiating epitheliochorial placentation. To analyze the transcriptome changes in the endometrium in the course of maternal recognition of pregnancy (MRP), deep sequencing of endometrial RNA samples of Day 12 pregnant animals (n=4) and corresponding non-pregnant controls (n=4) was performed using Illumina RNA-Seq. more...

Expression data from estrogen disrupted endometrium

(Submitter supplied) Placentation of the conceptus to the surface epithelium is governed through a tightly regulated temporal and spatial window. Premature exogenous steriod exposure causes a shift in the maternal tissue's receptivity and prevents proper placentation. We used Affymetrix microarrays to detail the aberrant changes in uterine gene expression during this window, while removing all conceptus contribution to this dysfunction.

Genome Wide Gene Expression Study of Circulating Monocytes in human with extremely high vs. low bone mass

(Submitter supplied) Peak bone mass (PBM) is an important determinant of osteoporosis. Circulating monocytes may serve as early progenitors of osteoclasts and produce important molecules for bone metabolism. To search for genes functionally important for osteoclastogenesis, we performed a whole genome gene differential expression study of circulating monocytes in human subjects with extremely low vs. high peak bone mass. more...

Expression data for stress treatment in rice seedlings

(Submitter supplied) Drought, salinity and sub-optimal temperatures are stresses that cause adverse effects on the growth of plants and the productivity of crops. In this study, we have analyzed the expression profiles of rice genes under control and abiotic stress conditions using microarray technology to identify the genes differentially expressed during various abiotic stresses. Expression data for reproductive development in rice

(Submitter supplied) Plants initially undergo a period of vegetative development, in which it produces leaves from shoot apical meristem (SAM) and roots from the root apical meristem. Later in development, the SAM undergoes a change in fate and enters reproductive development called as floral transition, producing flowers and seeds. Our understanding of the molecular and genetic mechanisms that underlie reproductive development in plants has increased tremendously in the past decade, essentially through the work on Arabidopsis. more...

Rice seedling hormone treatment

(Submitter supplied) Indica rice seedlings of IR64 variety were grown hydroponically for 7-days in a culture room with a daily photoperiodic cycle of 14h light and 10h dark. Seedlings were incubated in 0.1% dimethyl sulfoxide (control) or 50 micromolar solutions of indole-3-acetic acid (IAA treatment) and benzyl aminopurine (BAP treatment) for 1h and 3h. Equal amounts of 1h and 3h samoles were pooled for each treatment before RNA isolation. more...

Current smoking-specific gene expression signature in bronchial epithelium is enhanced in squamous cell lung cancer

(Submitter supplied) Cigarette smoking is the main risk factor for the development of squamous cell lung carcinoma (SCC). However, the smoking-related molecular changes in SCC have not been studied. We wanted to identify genes in both histologically normal bronchial epithelium and SCC samples that are differentially expressed between current and ex-smokers. In addition, to analyze the levels of the smoking-related genes identified in normal bronchial epithelium with the levels in SCC. more...

Identification of Differential Gene Expression During Transition of Bovine Corpus Luteum from Early To Mid–Phase.

(Submitter supplied) Prostaglandin F2 alpha (PGF2alpha) brings about regression of the bovine corpus luteum (CL). This luteolytic property of PGF2alpha is used in the beef and dairy cattle to synchronize estrus. A limitation of this protocol is an insensitivity of the early CL to luteolytic actions of PGF2alpha. The mechanisms underlying luteal sensitivity (LS) are poorly understood. The early CL has maximum number of PGF2alpha receptors; therefore differences in signaling events might be responsible for LS. more...

Understanding Bovine Embryo Elongation: A Transcriptomic Study of Trophoblastic Vesicles

(Submitter supplied) Background: During the process of elongation, the embryo increases in size within the uterus while the extra-embryonic tissues (EET) develop and differentiate in preparation for implantation. As it grows, the ovoid embryo changes shape into first a tubular and then a filamentous form. This process is directed by numerous genes and pathways, the expression of which may be altered in the case of developmental irregularities such as when the conceptus is shorter than expected or when the embryo develops after splitting. more...
