Murine 3T3-L1 adipocytes vs. adipocyte-derived microvesicles (ADMs)

(Submitter supplied) mRNA profiles of adipocyte-derived microvesicles (ADMs) and their donor 3T3-L1 adipoyctes (Day 11) were compared. ADMs included RNA without typical 28S and 18S ribosomal RNA.

Differentiation of NIH-3T3 cells to adipocytes by PPARg or EBF1 over-expression.

(Submitter supplied) NIH-3T3 cells transduced with either EBF1-, PPARg2- or empty vector were stimulated with hormones to initiate adipocyte differentiation. RNA extraction was done using TriZol at d0, d2, d4 and d10 after stimulation. Samples were handled according to standard affymetrix protocols. Fibroblast adipogenesis induced by EBF-1 and PPARgamma2 overexpression: time course

Analysis of NIH-3T3 embryonic fibroblasts induced to differentiate into adipocytes by overexpression of early B cell factor (EBF)-1 or peroxisome proliferator-activated receptor (PPAR)gamma2 transcription factors. Cells examined at various time points up to 10 days following overexpression.

A next generation sequencing based approach to identify extracellular vesicle mediated mRNA transfers between cells

(Submitter supplied) Purpose: Exosomes and other extracellular vesicles (EVs) have emerged as an important mechanism of cell-to-cell communication. In this work, we present a novel next generation of sequencing (NGS) based approach to identify EV mediated mRNA exchanges between co-cultured adipocyte and macrophage cells. Methods:We performed molecular and genomic profiling and jointly considered data from RNA sequencing (RNA-seq) and genotyping to track the ‘sequence varying mRNAs’ transferred between cells. more...

Glioblastoma microvesicles transport RNA and proteins

(Submitter supplied) Glioblastoma tumour cells release microvesicles (exosomes) containing mRNA, miRNA and angiogenic proteins. These microvesicles are taken up by normal host cells, such as brain microvascular endothelial cells. By incorporating an mRNA for a reporter protein into these microvesicles, we demonstrate that messages delivered by microvesicles are translated by recipient cells. These microvesicles are also enriched in angiogenic proteins and stimulate tubule formation by endothelial cells. more...
