Pulmonary alveolar macrophages (PAMs) of Tongcheng piglets response to HP-PRRSV infection in vivo

(Submitter supplied) Porcine reproductive and respiratory syndrome (PRRS), caused by PRRS virus (PRRSV), is the most economically important disease in pig populations. Lung damage is one major pathological condition following PRRSV infection, often leading to animal death. In vivo, PRRSV productive infection occurs predominately in alveolar macrophages of the lung. Here, transcriptome profiling of pulmonary alveolar macrophages (PAMs) from Tongcheng piglets pre- and post- infection of highly pathogenic PRRSV has been performed using porcine Affymetrix GeneChip.

Transcriptome analysis of the mRNAs and lncRNAs in porcine alveolar macrophages in response to porcine reproductive and respiratory syndrome virus or IFN-a at different time points

(Submitter supplied) Purpose:We performed next-generation RNA sequencing and a comprehensive bioinformatics analyses to characterize the transcriptome landscapes, including mRNAs and long noncoding RNAs (lncRNAs) in PRRSV-infected PAMs or IFN-stimulated PAMs at different time points. Conclusions: We showed that PRRSV infection displays different transcriptome profiles of PAMs, and the aberrant function of PAMs may be reflected by significant down-regulation of essential genes at the transcriptome levels. more...

Transcriptional profiling reveals differential effect of in vivo porcine reproductive and respiratory syndrome virus infection on infected and bystander pulmonary alveolar macrophage cells

(Submitter supplied) Porcine reproductive and respiratory syndrome virus (PRRSV) remains a serious threat to the swine industry worldwide for many years. PRRSV has a high tropism for swine pulmonary alveolar macrophages (PAM), an essential innate immune cell. Here, we used RNA-sequencing approach to examine the transcriptional profile of in vivo infected and bystander PAM in the lung of PRRSV-infected swine at 7 days post-infection (dpi). more...

Understanding normal PRRSV infection in lung based on genome-wide transcriptome response identified by deep sequencing

(Submitter supplied) Porcine reproductive and respiratory syndrome (PRRS) has been one of the most economically important diseases affecting swine industry worldwide and causes great economic losses each year. PRRS virus (PRRSV) replicates mainly in porcine alveolar macrophages (PAMs) and dendritic cells (DCs) and develops persistent infections, antibody-dependent enhancement (ADE), interstitial pneumonia and immunosuppression. more...

Genome-wide gene expression profiles in lung tissues of pig breeds differing in resistance to porcine reproductive and respiratory syndrome virus

(Submitter supplied) Porcine reproductive and respiratory syndrome caused by porcine reproductive and respiratory syndrome virus (PRRSV) is an infectious disease characterized by severe reproductive deficiency in pregnant sows, respiratory symptoms in piglets, and high mortality. In this study, we employed Affymetrix microarray chip technology to compare the gene expression profiles of lung tissue samples from Dapulian (DPL) pigs (a Chinese indigenous pig breed) and Duroc×Landrace×Yorkshire (DLY) pigs after infection with PRRSV. more...

Gene expression profiles for porcine alveolar macrophage isolated from piglets injected with Mab followed by challenge with two PRRSV strains: PRRSV-JXA1 and PRRSV-HNhx

(Submitter supplied) An PRRSV specific IgG-type Mab-PN9cx3, demonstrated broad recognition and neutralization activity against both PRRSV-1 and PRRSV-2 isolates in vitro, was tested for in vivo protection experiments. Administration of Mab-PN9cx3 20mg in piglets significantly alleviated the pathological changes in lungs and decreased virus loads in PAMs after challenging with two heterogenous PRRSV isolates: HP-PRRSV-JXA1 and PRRSV NADC-30 like HNhx when compared with piglets inoculated virus . more...

Whole Transcriptome analysis for porcine alveolar macrophage infected by African Swine Fever Virus

(Submitter supplied) African swine fever virus (ASFV) is one of the most devastating swine pathogens characterized by nearly 100% mortality in naive herds and was recently emerged the in China. In this study, we generated the expression profile of porcine alveolar macrophages (PAMs) infected with a high pathogenic ASFV (Pig/Heilongjiang/2018 (Pig/HLJ/18) ASFV). Our data indicated that ASFV infection lead to a strong inhibition of host immunity but promote chemokine-mediated signaling pathway and neutrophil chemotaxis. more...

PRRSV-infection of porcine alveolar macrophages

(Submitter supplied) The goal of this study was to produce a deep, global analysis of gene expression changes that occured following infection of normal porcine alveolar macrophages (PAMs) with PRRSV. The goal was to examine the gene expression changes to help determine the mechanisms that result in reduced function and immunosuppression observed in PRRSV-infected pigs. Keywords: time course of infection
