Human Beta-Defensin 3 attenuates the pro-inflammatory effects of LPS at 1 hour

(Submitter supplied) We examine the global effect of hBD3 on transcription in TLR4-stimulated macrophages and for the first time show that hBD3 inhibits the transcription of critical pro-inflammatory genes. Among the repressed genes we detect significant enrichment of groups involved in the positive regulation of NFkappaB including components of Toll-like receptor signaling pathways.

Gene Expression data from LPS (2 hour) - treated PPP2CA alpha conditional knockout Bone Marrow Derived Macrophages (PP2ACαfl/fl;lyM-Cre, cre-loxp mediated exon1 deletion in myeloid cells)

(Submitter supplied) PP2A regulates inflammatory cytokine/chemokine gene expression by dephosphorylating protein kinases at multiple signaling pathways from stimulated cells. In this dataset, Affymetrix mouse Gene ST 2.1 Array was used to assay total RNA extracted from LPS-treated PP2ACα knockout BMDM (PP2ACαfl/fl;lyM-Cre) and the control BMDM (PP2ACαfl/fl) In this dataset, we include the expression data obtained from LPS-stimulated PP2ACα conditional knockout BMDM (PP2ACαfl/fl;lyM-Cre) and control BMDM (PP2ACαfl/fl). more...

miRNA expression signatures in human alveolar macrophages after toll-like receptor activation

(Submitter supplied) We have employed miRNA microarray expression profiling to identify miRNAs that are differentially regulated by toll-like receptor activation in alveolar macrophages. Macrophages are strategically distributed all over the body and represent the first line of defense to invading pathogens. In response to bacterial or viral infections, these phagocytic cells engulf and destroy the infectious agent. Macrophages recognize a variety of microbial products, such as bacterial cell wall components and nucleic acids, via pathogen recognition receptors (PRRs), such as toll-like receptors (TLRs). more...

Mouse ischemic tolerance genomic analysis of the brain and blood.

(Submitter supplied) Ischemic tolerance can be induced by numerous preconditioning stimuli, including various Toll-like receptor (TLR) ligands. We have shown previously that systemic administration of the TLR4 ligand, lipopolysaccharide (LPS) or the TLR9 ligand, unmethylated CpG ODNs prior to transient brain ischemia in mice confers substantial protection against ischemic damage. To elucidate the molecular mechanisms of preconditioning, we compared brain and blood genomic profiles in response to preconditioning with these TLR ligands and to preconditioning via exposure to brief ischemia. more...
