The role of FGF21 in aging and aging-related diseases

(Submitter supplied) Aging and aging-related diseases represent an increasing burden on modern society. Thus, drugs that retard the aging process are highly desirable. Fibroblast growth factor-21 (FGF21) is a hormone secreted by the liver during fasting that elicits diverse aspects of the adaptive starvation response. Among its effects, FGF21 induces hepatic fatty acid oxidation and ketogenesis, increases insulin sensitivity and blocks somatic growth. more...

The effects of SGLT-2 inhibitors canagliflozin on hepatic transcription

(Submitter supplied) We assessed the change in hepatic transciptional pattern after treatment with SGLT-2 inhibitors canagliflozin in a mice model of diet-induced obesity. Pharmacologic inhibition of the renal sodium/glucose cotransporter-2 induces glycosuria and reduces glycemia. Given that SGLT2 inhibitors (SGLT2i) reduce mortality and CV risk in T2D, improved understanding of molecular mechanisms mediating these metabolic effects is required. more...

Gene expression in liver tissue from Ghrh-KO and normal (wild-type) mice

(Submitter supplied) The hypothalamus has recently emerged as a key regulator of metabolism and aging in mammals. We have examined the impact of targeted disruption of hypothalamic hypophysiotropic peptide: Growth Hormone-releasing Hormone (GHRH) in mice on longevity, and the putative mechanisms of delayed aging. GHRH knockout (KO) mice are remarkably long-lived and in comparison to genetically normal (wild type) animals exhibiting major shifts in the expression of genes related to xenobiotic detoxification, stress resistance, and insulin signaling. more...

Expression data from mouse liver after chow and high-fat diet feeding

(Submitter supplied) Epigenetic changes are present in multiple tissues of rodents and subjects suffering from obesity and type 2 diabetes. Especially, genes that are important for glucose and lipid metabolism seem to be at least co-regulated by DNA methylation. We aimed to investigate if altered DNA methylation in liver of mice can be induced by feeding a high-fat diet and if these changes can be reverted by weight loss. more...

Triiodothyronine (T3) response in liver of FGF21 knockout mice

(Submitter supplied) Thyroid hormone (TH) influences metabolic pathways by binding to specific receptors (TRs), which are conditional transcription factors. T3 works through TRs to induce fibroblast growth factor (FGF) 21, a peptide hormone that is usually induced in fasting and influences lipid and carbohydrate metabolism via local hepatic and systemic endocrine effects. While administered TH and FGF21 display overlapping actions, including reductions in serum lipids, current models suggest that these hormones act independently in vivo. more...
