Gene expression analysis in macrophages stimulated 18 h with Adiponectin

(Submitter supplied) Bone marrow derived macrophages (BMDM) generated from c57bl/6j mice bone marrow cells were stimulated for 18 h with 12 microgram/ml adiponectin, RNA from non-stimulated or 18 h adiponectin-stimulated BMDM subjected to a agilent microarray analysis

Tpl2 promotes chemokine/chemokine receptor expression and macrophage migration during acute inflammation

(Submitter supplied) In autoimmune diseases, accumulation of activated leukocytes correlates with inflammation and disease progression, and therefore, disruption of leukocyte trafficking is an active area of research. The protein kinase Tpl2 (MAP3K8) regulates leukocyte inflammatory responses and is also being investigated for therapeutic inhibition during autoimmunity. Herein, we addressed the contribution of Tpl2 to the regulation of macrophage chemokine and chemokine receptor expression and subsequent migration in vivo using a mouse model of Tpl2 ablation. more...

Lipopolysaccharide effect on tumor progression locus 2 kinase-deficient macrophages

Analysis of tumor progression locus 2 (Tpl2)-deficient bone marrow derived macrophages (BMDMs) stimulated with lipopolysaccharide (LPS) for 4 hrs. Tpl2 (Map3k8) is a serine/threonine kinase critical in innate immunity. Results provide insight into the role of Tpl2 in BMDMs during acute inflammation.

Transcriptomic analysis of IRAK4 in murine pancreatic cancer cells (KP2)

(Submitter supplied) We perform CRISPR knockout of IRAK4 usig two different sgRNAs in murine KP2 cells, and then reexpressed murine IRAK4 in these two KO cell lines. We performed RNAseq on wild-type, IRAK4 KO and IRAK4 KO/rescue cell lines to investigate pathways contolled by IRAK4.

Ferret animal model of severe fever with thrombocytopenia syndrome phlebovirus for human lethal infection and pathogenesis

(Submitter supplied) Severe fever with thrombocytopenia syndrome phlebovirus (SFTSV), listed in the WHO most dangerous pathogens, has 12-30% fatality rates with a characteristic thrombocytopenia syndrome. With a majority of clinically diagnosed SFTSV patients older than ~50 years, age is a critical risk factor for SFTSV morbidity and mortality. Here, we report an age-dependent ferret model of SFTSV infection and pathogenesis that fully recapitulates the clinical manifestations of human infections. more...

The matricellular protein CCN1 activates a pro-inflammatory genetic program in murine macrophages

(Submitter supplied) Effects of the matricellular protein CCN1 on macrophage inflammatory gene expression was tested in a macrophage cell line I-13.35. CCN1 (CYR61) is a matricellular protein that is highly expressed at sites of inflammation and wound repair. In these contexts, CCN1 can modify the activities of specific cytokines, enabling TNF-alpha to be cytotoxic without blocking NFkB activity and enhancing the apoptotic activity of FasL and TRAIL. more...

Expression data from human PBMC samples exposed to IL-12 and paired controls

(Submitter supplied) The cytokine interleukin-12 (IL-12) is known to play a central role in adaptive and innate immunity. We employed microarray analysis of IL-12 induced gene expression to provide further insights into its effects on immune response. Keywords: Paired study of expression levels: Treated vs. Control
