Comparative Transcriptomics of Choroid Plexus in Alzheimer's Disease, Huntington's Disease and Frontotemporal Dementia: Implications for CSF Homeostasis and Dynamics

(Submitter supplied) Background: In aging and Alzheimer’s disease there are striking changes in CSF composition that may relate to altered choroid plexus function. Studying CP tissue gene expression at the BCSFB provides further insight into epithelial and stromal responses to aging and diseased states. Results: Highly significant differences in gene expression occurred in CP of advanced AD patients vs. normal controls. more...

Gene expression data from lateral ventricle choroid plexuses of developing and adult rats

(Submitter supplied) Choroid plexuses (CP) develop early during development. They form a barrier between the blood and the cerebrospinal fluid, and fulfill important protective and nutritive functions. We used Affymetrix microarrays to assess whether CP of the lateral ventricles (LVCP) have similar functions in developing and adult brain. We identified distinct families of protective and transport genes and found that most of these genes were already well expressed during development.

Comparison of mouse and human retinal pigment epithelium gene expression profiles reveals interspecies similarities and differences

(Submitter supplied) To determine mouse retinal pigment epithelium (RPE) specific gene expression, we compared the gene expression profile of the RPE to the gene expression of its adjacent layers: the choroid (CH) and photoreceptors (PR). We collected the tissues using the meticulous laser dissection microscopy. Next, we performed RNA isolation, amplification, labeling and hybridization against 44k Agilent microarrays. Compared RPE gene expression with CH gene expression, and with PR gene expression

OTX2 homeoprotein regulates adult choroid plexus activity

(Submitter supplied) Choroid plexus secretes cerebrospinal fluid important for brain development and homeostasis. The OTX2 homeoprotein is critical for choroid plexus development and remains highly expressed in adult choroid plexus. Through RNA sequencing analyses of constitutive and conditional knockdown adult mouse models, we reveal putative roles for OTX2 in choroid plexus function, including cell signaling and adhesion, and show that it regulates the expression of factors secreted into cerebrospinal fluid, notably transthyretin. more...

Spatially heterogeneous choroid plexus transcriptomes encode positional identity and contribute to regional cerebrospinal fluid production

(Submitter supplied) A sheet of choroid plexus epithelial cells extends into each cerebral ventricle and secretes signaling factors into the cerebrospinal fluid (CSF). To evaluate whether differences in the CSF proteome across ventricles arise, in part, from regional differences in choroid plexus gene expression, we defined the transcriptome of lateral ventricle (telencephalic) vs. fourth ventricle (hindbrain) choroid plexus. more...

A cellular and spatial map of the choroid plexus across brain ventricles and ages

(Submitter supplied) The choroid plexus (ChP) in each brain ventricle produces cerebrospinal fluid (CSF) and forms the blood-CSF barrier. We apply single cell and single nuclei sequencing to identify region and age specific shifts in gene expression of the constituent cell types of the choroid plexus. First, we sequenced whole cells (15,620) from each ventricle (lateral, third and fourth) of the embryonic mouse brain (Embryonic day (E) 16.5). more...

The choroid plexus transcriptome reveals changes in type I and II interferon responses in a mouse model of Alzheimer's disease

(Submitter supplied) Alzheimer’s disease (AD) is a neurodegenerative disease characterized by a marked decline in cognition and memory formation. Increasing evidence highlights the essential role of neuroinflammatory and immune-related molecules, namely the ones that are produced at the brain barriers, on brain immune surveillance, cellular dysfunction and amyloid beta (Aβ) pathology in AD. Therefore, understanding the response at the brain barriers may unravel novel pathways that are relevant for the pathophysiology of AD. more...
