Systems level analyses establish CodY as a multifaceted metabolic regulator integrating metabolism, motility and virulence in Listeria monocytogenes [ChIP-Seq]

(Submitter supplied) To gain knowledge on L. monocytogenes differential CodY activity, we set on analyzing CodY’s regulon in L. monocytogenes in both rich and minimal growth conditions using genome wide sequencing techniques. Remarkably, we identified for the first time a global regulatory role for CodY when BCAAs are limited, that are similar to those within the mammalian niche. Furthermore, our data establish CodY as a central regulator that integrates metabolism, motility, stress responses and virulence in L. more...

RNA-sequencing based analysis of Listeria monocytogenes 10403S::ΔBCHL Prha-sigH and ΔBCHL Prha

(Submitter supplied) In this study, RNA-seq was used to compare the transcriptomes of Listeria monocytogenes 10403S::ΔBCHL Prha-sigH and ΔBCHL Prha. RNA-seq was performed on ΔBCHL Prha-sigH and ΔBCHL Prha RNA samples representing three independent biological replicates at log phase in Brain Heart Infusion (BHI) broth under rhamnose induction. Indexed and purified cDNA libraries (6 libraries including 3 replicates for 2 strains) were loaded together onto an independent flow cell without any other samples; sequencing was carried out by running Hiseq 2500 (single-end, 150-bp per read). more...

Adaption of Dinoroseobacter shibae to anaerobic conditions

(Submitter supplied) To unravel the adaptation strategies of D. shibae to anaerobic conditions in microaerobic to anaerobic parts of the ocean and to define the underlying regulatory network an anaerobic shift experiment in Salt-Water-Medium in a chemostate was established. Transcriptome analyses were used to investigate the physiological status of D. shibae under this conditions.

Genome-wide transcriptional profiling of the cell envelope stress response of Listeria monocytogenes

(Submitter supplied) The Gram-positive bacterium Listeria monocytogenes is widely distributed in the environment and capable of causing food-borne infections in susceptible individuals. In this study, we investigated the cell envelope stress response in L. monocytogenes. Whole-genome transcriptional profiling was performed to investigate the response upon exposure to the cell wall antibiotic cefuroxime. Differential expression (≥ 2-fold difference) of 558 genes was observed, corresponding to 20% of the L. more...

Exploring Listeria monocytogenes transcriptomes in correlation with divergence of lineages and virulence measured in Galleria mellonella

(Submitter supplied) For bacteria in general and Listeria monocytogenes in particular, little is known about transcriptome diversity in comparison to the copious data on intraspecific genome diversity. The current study employed RNA sequencing to investigate the variation of transcript levels of conserved single copy genes from several perspectives including differences between lineages and correlation with previous classification of the virulence potential of clonal complexes based on epidemiological MLST data. more...
