Genome-wide analysis of histone modifications in human endometrial stromal cells [RNA-Seq]

(Submitter supplied) Dramatic changes of gene expressions are known to occur in human endometrial stromal cells (ESC) during decidualization. The changes in gene expression are associated with changes of chromatin structure, which are regulated by epigenetic mechanisms such as histone modifications. Here, we investigated genome-wide changes in histone modifications and mRNA expressions associated with decidualization in human ESC using chromatin immunoprecipitation (ChIP) combined with next-generation sequencing. more...

Expression data from non-decidualized and decidualized endometrial stromal cell (ESCs) with or without WT1 knockdown

(Submitter supplied) Wilms tumor 1 (WT1) regulates the expression of insulin-like growth factor-binding protein-1 (IGFBP-1) and prolactin (PRL) during decidualization of human endometrial stromal cells (ESCs). However, other roles of WT1 in decidualization have not been fully clarified. We used microarrays to identify the physiological roles of WT1 in the regulation of cellular functions of human ESCs during decidualization.

Genome-wide Profiling of NR2F2 (COUP-TFII) Binding in Human Endometrial Stromal Cells [ChIP-Seq]

(Submitter supplied) Chicken ovalbumin upstream promoter-transcription factor II (COUP-TFII; NR2F2) is an orphan nuclear receptor involved in cell-fate specification, organogenesis, angiogenesis and metabolism. Ablation of COUP-TFII in the mouse uterus causes infertility due to defects in embryo attachment and impaired uterine stromal cell decidualization. Although the function of COUP-TFII in uterine decidualization has been described in mice, its role in the human uterus remains unknown. more...

Homeobox regulator Wilms tumour 1 (WT1) is displaced by androgen receptor at cis-regulatory sites in the endometrium of PCOS patients.

(Submitter supplied) Polycystic ovary syndrome (PCOS) is a common gynaecological disorder, characterised by elevated circulating androgens and increased expression of androgen receptor (AR) and by downregulation of Wilms Tumour 1 in the endometrium, The objectives of this study are: 1) Understand the functional role of WT1 in regulating decidualization response in healthy endometrial tissue, and 2) understand how interactions between aberrant WT1 and AR expression in PCOS patients result in dysregulation of deciduallization processes

H3K4me3 and H3K9me3 histone modification profiles of uterine stromal cells from young and aged mouse females

(Submitter supplied) Epigenetic profiling of uterine stromal cells (UtSCs) was performed for the active and repressive histone modifications H3K4me3 and H3K9me3, respectively. We find that UtSCs from aged females are epigenetically distinct, as determined by genomic enrichment profiling for H3K4me3 and H3K9me3, respectively. We find that many pregnancy hormone-inducible genes exhibit a profound lack of promoter-associated H3K4me3 in aged UtSCs, implying that a significant enrichment of active histone marks prior to gene stimulation is required to enable the elicitation of a rapid transcriptional response.

Expression data from in vitro decidualized human uterine fibroblast cells (HuF cells) with or without NOTCH1 silencing

(Submitter supplied) Decidualization is a critical process for embryo implatation during which uterine stromal fibroblasts are transformed into large, epithelioid-like decidual cell. NOTCH1 is recepotor of Notch signaling that plays important roles for cell-cell communication, which involves gene regulatory mechanisms that control multiple cellular differentiation processes during embryonic and adult life. Here we silenced NOTCH1 by shRNA in HuF cells and studied how it affects decidualization

Genome-wide analysis of the rat colon reveals site-specific differences in histone modifications and proto-oncogene expression

(Submitter supplied) Goal: Since disease susceptibility of the intestine exhibits an anatomical bias, we propose that the chromatin landscape, especially the site-specific epigenetic differences in histone modification patterns throughout the longitudinal axis, contributes to a differential response to chemoprotective agents. Method: We assessed the chromatin structure associated with gene expression profiles in the rat proximal and distal colon by globally correlating chromatin immunoprecipitation next-generation sequencing analysis (ChIP-Seq) with mRNA transcription (RNA-Seq) data. more...
