Addiction to the IGF2-ID1-IGF2 circuit for maintenance of the breast cancer stem-like cells.

(Submitter supplied) The transcription factor nuclear factor-κB (NF-κB) has important roles for tumorigenesis, but how it regulates cancer stem cells (CSCs) remains largely unclear. We identified insulin-like growth factor 2 (IGF2) is a key target of NF-κB activated by HER2/HER3 signaling to form tumor spheres in breast cancer cells. The IGF2 receptor, IGF1 R, was expressed at high levels in CSC-enriched populations in primary breast cancer cells. more...

Crosstalk between the STAT3, NF-kB and Notch signaling pathways in glioblastoma stem cells

(Submitter supplied) In this study, we describe a novel relationship between glioblastoma CSCs and the Notch pathway, which involves the constitutive activation of STAT3 and NF-κB signaling. We demonstrate that adherent glioma CSCs exhibit characteristics previously described for CSCs grown in suspension culture. The expression of CD133, Sox2 and Nestin increased when compared to glioma cells grown in monolayer, and the adherent CSCs were ~100 times more tumorigenic in vivo than monolayer cultured glioma cells. more...

Naa10p's effect on gene expression profiles in MCF7 breast cancer cells

(Submitter supplied) N-α-acetyltransferase 10 protein (Naa10p, also called ARD1), the catalytic subunit of N-acetyltransferase A, is a critical regulator of cell death and proliferation. Naa10p is also shown to regulate cancer metastasis by inhibiting cell motility, however its role in cancer metastasis is not fully understood. In this study, we found that high expression of Naa10p is positively correlated with the survival of patients with breast cancer, while negatively correlated with lymph node metastasisr. more...

Gene expression of clinical breast cancer cells cultured by sphere and adherent culture methods

(Submitter supplied) Cancer stem-like cells (CSCs) are thought to be responsible for recurrence of tumors and poor prognosis because of their higher tumor initiating ability and drug resistance, though the mechanisms remain still unclear. Here, we show a critical role of mini-chromosome maintenance protein (MCM) 10, a component of DNA replication machinery, in enabling CSCs to deal with DNA replicative stress. Our transcriptomic analysis of patient-derived cultured breast cancer cells revealed that MCM10 is strongly upregulated in CSC-enriched spheroid cells, compared to cancer cells cultured in a regular condition. more...

A SIRT1-centered Circuitry Regulates Breast Cancer Stemness and Metastasis

(Submitter supplied) Deficiency of deacetylase SIRT1 or its downstream target PRRX1 promotes breast cancer stemness and metastasis. S100A9 is a mediator of the ID1 functions in metastasis

(Submitter supplied) Metastasis is a major factor responsible for mortality in breast cancer patients. Id1 plays important roles in cell differentiation and promotes tumor angiogenesis, cell invasion and metastasis. Although Id1 is established as a critical factor for lung metastasis in breast cancer, the pathways and molecular mechanisms of Id1 functions in metastasis remains to be defined. Here we show that Id1 interacts with TFAP2A to suppress S100A9 expression. more...

A facile method for generating tumorigenic cancer stem cell spheroids from diverse cancer cell lines

(Submitter supplied) Although cancer stem cells (CSCs) are thought to be responsible for tumor recurrence and resistance to chemotherapy, CSC-related research and drug development have been hampered by the limited supply of patient-derived diverse CSCs. Here, we developed a functional polymer thin film (PTF) platform that promotes conversion of human cancer cell lines to highly tumorigenic spheroids without the use of biochemical or genetic manipulations. more...

Expression Data from ALDH1+ breast cancer stem cells

(Submitter supplied) Breast cancer is the most common cancer in women worldwide and metastatic dissemination is the principal factor related to death by this disease. Breast cancer stem cells, are thought to be responsible for metastasis and chemoresistance.. In this study, based on whole transcriptome analysis from putative breast CSCs and reverse-engineering of transcription control networks, we were able to identify two networks associated to this phenotype.
