Comparison of miRNA expression in HT29-tsp53 cells cultured at the permissive and restrictive temperatures

(Submitter supplied) The p53 tumour suppressor is a transcription factor that can regulate the expression of numerous genes encoding either proteins or microRNAs (miRNAs). The predominant outcomes of a typical p53 response are the initiation of apoptotic cascades and the activation of cell cycle checkpoints. HT29-tsp53 cells express a temperature sensitive variant of p53 and in the absence of exogenous DNA damage, these cells preferentially undergo G1 phase cell cycle arrest at the permissive temperature that correlates with increased expression of the cyclin-dependent kinase inhibitor p21WAF1. more...

Expression profiling of lncRNAs, miRNAs and mRNAs and their differential expression in leiomyoma using next generation RNA sequencing

(Submitter supplied) Total RNA was isolated from leiomyoma and paired myometrium (N=8) and samples from three pairs were subjected to RNA sequencing. After analysis, 5941 lncRNAs (2813 up- and 3128 down-regulated at ≥1.5 fold), 148 miRNAs (56 up- and 96 down-regulated at ≥1.5 fold) and 3855 mRNAs (2030 up- and 1855 down-regulated at ≥1.5 fold) were differentially expressed in leiomyomas. Using QRT-PCR we further confirmed the expression of HULC, lnc-MEG3, LINC00890, TSIX, LINC00473, lnc-KLF9-1 and lnc-POTEM-3 (lncRNA-ATB) in leiomyoma and matched myometrium (N=8). more...

Expression data from human embryonic stem cell differentiation

(Submitter supplied) MicroRNAs (miRNAs) are noncoding RNAs of approximately 22 nucleotides in length that usually suppress the translation of target messenger RNAs (mRNAs) through partial complementarity to the 3¡¦ untranslated region (3¡¦ UTR) of protein-coding mRNAs in animals. However, there is increasing evidence that miRNAs can also reduce the steady-state levels of their target mRNAs in animals. In this investigation, both miRNA and mRNA profiles from the undifferentiated human embryonic stem cell line hES-T3 (T3ES), hES-T3 derived embryoid bodies (T3EB) and hES-T3 differentiated fibroblast-like cells (T3DF) were quantitatively determined. more...

smRNA sequencing analysis to identify p53-dependent non-coding RNA networks in Chronic Lymphocytic Leukemia

(Submitter supplied) Mutations of the tumor suppressor p53 lead to chemotherapy resistance and a dismal prognosis in chronic lymphocytic leukemia (CLL). Comparing miRNA/non-coding RNA expression profiles between p53 wild-type and p53 mutant samples in response to DNA damage, we exploit the impaired transcriptional activity of mutant p53 to map out p53 targets in primary CLL by small RNA sequencing. Focusing on miRNAs, we identify a set of p53-dependent miRNAs (miR-182-5p, miR-7-5p, miR-320c/d) in addition to the key p53 target miR-34a. more...

(Submitter supplied) We report the application of high-throughput sequencing to performed the p53 regulated trancriptome in HCT116 colon cancer cells treated with the DNA damage 5FU. To study the direct targets of p53 we performed ChIP-seq to deterrmined the p53 biding sites and associated with the expression levels. With this study we identified the new genomic regions regulated by p53 and with special attention in those regions that are non coding and are differentially expressed by the DNA damage drug.

Identification of human miR-34a-responsive transcripts

(Submitter supplied) In order to examine the consequences of human miR-34a induction on the transcriptome, HCT116 cells (a colon cancer cell line) were infected with a retrovirus that produces miR-34a. Gene expression profiles were then monitored using Affymetrix microarrays. Affymetrix microarrays were used to examine the transcriptomes of HCT116 cells infected with an empty retroviral vector (pMSCV-PIG) or a retroviral vector that expresses human miR-34a. more...
