Aflatoxin B1 induces persistent epigenomic effects in primary human hepatocytes associated with hepatocellular carcinoma [miRNA]

(Submitter supplied) Chronic exposure to aflatoxin B1 (AFB1) has, in certain regions in the world, been strongly associated with the development of hepatocellular carcinoma (HCC). AFB1 is a very potent hepatotoxic and carcinogenic mycotoxin which is frequently reported as a food contaminant. Epigenetic modifications provoked by environmental exposures, such as AFB1, may create a so called persistent "epigenetic memory" or "footprint". more...

Effect of the genotoxic human lung carcinogen benzo[a]pyrene and its non-carcinogenic isomer benzo[e]pyrene on transcriptomic profiles in HepaRG cells.

(Submitter supplied) Twenty-eight days after the initial seeding, the terminally differentiated HepaRG cells were treated with 2 µM benzo[a]pyrene (B[a]P) and benzo[e]pyrene (B[e]P) for 72 hours. Following the treatment, the cells were harvested by mild trypsinization, washed in phosphate-buffered saline, and immediately frozen at −80°C for subsequent analyses. Gene expression profiles in the HepaRG cells treated with B[a]P and B[e]P were investigated using Agilent whole genome 8x60K human microarrays according to the manufacturer’s instructions.

Gene expression in trout hepatocellular carcinoma

(Submitter supplied) Hepatocellular carcinoma (HCC) is one of the most common malignant tumors worldwide and its occurrence is associated with a number of environmental factors including ingestion of the dietary contaminant aflatoxin B1 (AFB1). While etiology of the disease is relatively well defined, the molecular mechanisms of tumorigenesis are poorly understood. In this study, we examined, for the first time, gene expression of AFB1-induced HCC by microarray in the rainbow trout (Oncorhynchus mykiss) tumor model. more...

Genome-wide promoter DNA Methylation Profiling of Hepatocellular Carcinomas Arising Either Spontaneously or due to Chronic Exposure to Ginkgo biloba Extract (GBE) in B6C3F1/N Mice [Methylation]

(Submitter supplied) Epigenetic modifications, such as DNA methylation, play an important role in carcinogenesis. In a recent NTP study, chronic exposure of B6C3F1/N mice to Ginkgo biloba extract (GBE) resulted in a high incidence of hepatocellular carcinomas (HCC). Genome-wide promoter methylation profiling on GBE-exposed HCC (2000 mg/kg group), spontaneous HCC (vehicle-control group) and age-matched vehicle control liver was performed to identify differentially methylated genes in GBE-exposed HCC and spontaneous HCC. more...

Integrated analysis of genomic and epigenomic regulation of transcriptome identifies molecular subtypes of liver cancer (gene expression)

(Submitter supplied) We profiled DNA copy numbers, DNA methylation, and mRNA expression levels in the same HCC patients. To identify genes which transcriptional expression levels are regulated by genomic and/or epigenetic deregulation, we defined the copy number-correlated and methylation-correlated genes with their corresponding gene expression levels across the samples as genic and epigenetic deregulators, respectively.

Genetic and epigenetic characteristics of human multiple hepatocellular carcinoma

(Submitter supplied) Multiple carcinogenesis is one of the major characteristics of human hepatocellular carcinoma (HCC). The history of multiple tumors, i.e., whether they are derived from a common precancerous or cancerous ancestor or individually from hepatocytes, is a major issue. Multiple HCC is clinically classified into intratumor metastasis (IM) and multicentric carcinogenesis (MC). Molecular markers differentiating IM and MC are of interest to clinical practitioners because clinical diagnosis of IM and MC often leads to different therapies. more...
