Gene expression data from Pseudomonas aeruginosa PAO1 and mutator (Δ mutS) evolved for 940 generations in LB with and without sub-inhibitory concentrations of ciprofloxacin (0.05µg/ml)

(Submitter supplied) Gene expression of P. aerruginosa changes after short-term exposure to ciprofloxacin at sub-inhibitory concentrations but the effect of long-term exposure which select for the most fitted subpopulations is not known. We used microarrays to investigate the changes in gene expression of P.aeruginosa PAO1 and mutator (Δ mutS) after long-term evolution (94 daily passages) in LB in the presence and absence of ciprofloxacin

PAHM4 vs PAO1 37C LB

(Submitter supplied) Pseudomonas aeruginosa strains PAHM4 and PAO1 were grown at 37C on LB and RNA was hybridized on the Affymetrix P. aeruginosa chip to compare transcript differences from a BQ isolate to a well characterized wound isolate.

Pseduomonas aeruginosa PAO1 exposed to subinhibitory colistin

(Submitter supplied) Colistin is an important cationic antimicrobial peptide (CAMP) in the fight against Pseudomonas aeruginosa infection within the cystic fibrosis (CF) lungs. The effects of sub-inhibitory colistin on gene expression in P. aeruginosa were investigated by transcriptome microarray and functional analysis. Analysis revealed an alteration in the expression of 60 genes in total from a variety of pathways. Genes associated with bacterial chronic colonisation and virulence such as response to osmotic stress, motility, and biofilm formation, as well as those associated with LPS modification and quorum sensing are the most highly represented. Most striking among these is the upregulation of the PQS biosynthesis operon including pqsH, pqsE, and the anthranilate biosynthetic genes phnAB. Early activation of this central component of the QS-network may represent a switch to a more robust population, with increased fitness in the competitive environment of the CF-lung.

Pseduomonas aeruginosa PAO1 response to subinhibitory colistin

Analysis of Pseduomonas aeruginosa PAO1 treated with colistin at a subinhibitory concentration (0.15 ug/ml). Colistin is an antimicrobial peptide against P. aeruginosa infection in cystic fibrosis lungs. Result provide insight into the adaptive response of P. aeruginosa to low doses of colistin.

The polyaminoisoprenyl compound NV716 re-sensitizes antibiotics against Pseudomonas aeruginosa by perturbing outer membrane permeability and reduces bacterial virulence.

(Submitter supplied) Objective: to determine the changes in transcriptome induced by exposure during 52 successive passages of PAO1 to membrane-active agents, namely colistin (CST), alexidine (ALE) and NV716, an investigation polyamino-isoprenic compound Methods: PAO1 was exposed during 50 passages to each of these agents at 1/2 MIC, after which , cells were harvested, and RNA was purified and analyzed by RNAseq Results: upregulations of genes involved in lipidA/LPS synthesis as well as downrelgulation of genes involved in virulence were observed with NV716. more...
