Gene expression of young, middle-aged and old Drosophila melanogaster exposed to different levels of larval and adult diet

(Submitter supplied) Many studies have addressed the effects of adult diet on gene expression in Drosophila melanogaster, however, little is known about how developmental diet influences adult gene expression, and how this interacts with adult dietary conditions. We found that developmental and adult diet exert largely independent effects on gene expression, with the effect of adult diet being considerably larger. We did find effects of developmental diet on the transcriptome that persist into middle and old-age. more...

The effect of larval diet on the fecundity-longevity relationship and age-related gene expression in Drosophila melanogaster

(Submitter supplied) Our two main aims were 1) to isolate age-related changes in gene expression in females of the solitary insect, Drosophila melanogaster; and 2) to determine whether experimentally changing the shape of the fecundity-longevity relationship (by changing larvae diet) caused changes in age-related gene expression in these females. To address these aims we extracted RNA from three key tissues (fat body, head and ovary) from females at two time points (10% and 60% mortality phases). more...

Microarray analysis on fat bodies of fruit fly (Drosophila melanogaster) larvae fed with either a semi-purified diet only or a semi-purified diet supplemented with palm fruit juice (PFJ)

(Submitter supplied) PFJ (4 ml for a final concentration of 19,000 mg gallic acid equivalent (GAE) per kg diet or 0.86 mg GAE per kcal diet) was supplemented to larvae of fruit flies (Drosophila melanogaster) given a semi-purified diet to observe for possible effects on energy metabolism and lifespan. Fat bodies extracted from these larvae were used five days since the egg stage for gene expression studies. Results from the microarray data analysis carried out show that fruit fly larva fat bodies given PFJ had up-regulated heat shock protein genes, while cell cycle and growth genes were down-regulated.

Genotype-by-diet interaction effects on transcriptome profiles in Drosophila melanogaster

(Submitter supplied) This study was performed by testing 21 wildtype genetic lines on four different diets to look at the transcriptomic, metabolomic, and gross phenotype variation. The diets used were Normal Cornmeal-molasses symbolized N, high sugar symbolized 4, high fat F, and control C a low sugar diet. Larvae were raised from hatching to 3rd instar on the respecitve diets at a density of 50 larvae per food vial. more...

Diet-induced plasticity of life-history traits and gene expression in outbred Drosophila melanogaster population

(Submitter supplied) Food is a fundamental requirement for organisms to develop, survive and reproduce. However, access to an optimum diet is not always guaranteed, resulting in a mismatch between the diet and the organism's needs. To comprehend the consequences of this mismatch, it becomes crucial to explore how different compositions of an organism's diet impact its overall performance. In this context, we tested the effect of isocaloric diets with different protein to carbohydrate ratios on eight key life history traits in a large (n = ~2,100) outbred Drosophila melanogaster population. more...

Aging Fly Cell Atlas Identifies Exhaustive Aging Features at Cellular Resolution

(Submitter supplied) Aging is accompanied by the functional decline of all tissues, but it is still largely unknown how aging impacts different tissues in a cell type-specific manner. Here, we present the Aging Fly Cell Atlas (AFCA) that includes single-nucleus transcriptomes of the entire Drosophila head and body from both males and females at four different ages. We characterize 162 distinct cell types and present an in-depth analysis of cell type-specific aging features, including changes of cell composition, gene expression, number of expressed genes, transcriptome noise, and cell identity. more...
