A SRC-3 gene signature in predecidual T-HESC

(Submitter supplied) Steroid Receptor Coactivator-3 (SRC-3) knockdown in human endometrial stromal cells (HESCs) blocks their decidualization. This result provides translational support for recent studies in the mouse in which conditional SRC-3 knockout in progesterone receptor-positive cells of the endometrium results in early pregnancy loss due to a defect in normal decidualization. RNAseq was performed on the telomerase-immortalised endometrial stromal cell line T-HESC (CRL-4003; American Type Culture Collection) with or without SRC-3 knockdown to identify the transcriptome that is dependent on SRC-3 prior to hormone-dependent HESC decidualization.

Gene expression analyses of PR action in the uteri of SRC-2 mutant mice

(Submitter supplied) Ovarian estrogen (E2) and progesterone (P4) are indispensable for embryo-implantation and endometrial stromal decidualization; however, the molecular mechanisms that underpin these reproductive processes are unclear. Steroid receptor coregulator-2 (SRC-2) belongs to the multifunctional SRC/p160 family which also includes SRC-1 and SRC-3. Sharing strong sequence homology, all three SRCs exert diverse regulatory effects by modulating the transcriptional potency of nuclear receptor family members, including the estrogen and progesterone receptor (ER and PR respectively). more...

Progesterone effect on uteri of ovariectomized, steroid receptor coregulator-2 deficient mutants

Analysis of uteri from steroid receptor coregulator-2 (SRC-2) mutants that were ovariectomized at 6 weeks and treated with progesterone (P4) 2 weeks later. SRC-2 is a member of the p160/SRC family of coregulators. Results provide mechanistic insight into SRC-2's role in endometrial decidualization.

Genome-wide binding sites of Progesterone receptor (PGR) isoforms PGR-A and PGR-B during differentiation of human endometrial stromal cells

(Submitter supplied) We report the genome-wide binding sites of PGR-A and PGR-B at 2h of in vitro differentiation of human endometrial stromal cells that express either PGR-A or PGR-B. Progesterone, acting through the progesterone receptors (PGRs), is one of the most critical regulators of endometrial differentiation, known as decidualization, which is a key step toward the establishment of pregnancy. Yet a long-standing unresolved issue in uterine biology is the precise roles played by the major PGR isoforms, PGR-A and PGR-B, during decidualization in the human. more...

The coregulator, repressor of Estrogen Receptor Activity (REA), is a crucial regulator of the timing and magnitude of uterine decidualization.

(Submitter supplied) Our findings establish a key role for the coregulator, Repressor of Estrogen receptor Activity (REA), in controlling the timing and magnitude of decidualization in human endometrial stromal cells in vitro and in the mouse uterus in vivo, and suggest that REA functions to synchronize uterine differentiation with concurrent embryo development, which is essential for optimal implantation and fertility. more...

Expression data of differentially regulated genes in ESR1 depleted human endometrial stromal cells during differentiation.

(Submitter supplied) Estrogen and progesterone are important regulators of human endometrial differentiation. These steroid hormones act, at least in part, through their nucelar receptors. Role of estrogen receptor alpha (ESR1) during human endometrial differentiation is still unclear. We used microarray analysis to detail the gene expression regulated by ESR1 during differentiation of human endometrial stromal cells.

Steroid Receptor Coactivator-2 Drives Epithelial Reprogramming That Enalbles Murine Embryo Implantation

(Submitter supplied) Using conditional knockout technology, we demonstrate that steroid receptor coactivator-2 (SRC-2) is essential for development of the receptive uterus for embryo implantation. Through bulk RNA-seq analysis, we reveal that endometrial SRC-2 is required for not only the induction of the expression of established genes associated with uterine receptivity but also for the expression of genes involved in endometrial epithelial changes that enable attachment and invasion of the embryo into the underlying stroma. more...

Krüppel-like Factor 9 and Progesterone Receptor Co-Regulation of Decidualizing Endometrial Stromal Cells: Implications for its Loss in the Pathogenesis of Endometriosis

(Submitter supplied) Endometriosis is characterized by progesterone resistance and is associated with infertility. Krüppel-like Factor 9 (KLF9) is a progesterone receptor (PGR)-interacting protein, and mice null for Klf9 are subfertile. Whether loss of KLF9 contributes to progesterone resistance of eutopic endometrium of women with endometriosis is unclear. The aim of this study was to investigate KLF9 and PGR co-regulation of human endometrial stromal cell (HESC) transcriptome network.

SOX4 facilitates PGR protein stability and FOXO1 expression conducive for human endometrial decidualization

(Submitter supplied) To detect the direct target genes of SOX4 in differentiated human endometrial stromal cells(hEnSC), differentiated hEnSC cells at the presence of MPA, E2 and cAMP are collected and subjected to ChIP-Seq. After aligned to human Hg38 by STAR, peaks are called by MACS2. Our results show that SOX4 is the key regulator for decidualization by regulating FOXO1, STAT3, PRL and FOSL2. This specific enrichment is confirmed by ChIP-qPCR. more...
