FOXO and HSF1 rejuvenate immunity via positive feedback regulation of insulin/IGF-1 signaling

(Submitter supplied) A hallmark of aging is immunosenescence, a decline in immune function that appeared to be inevitable. Surprisingly, here we show that genetic inhibition of DAF-2/insulin/IGF-1 receptor drastically delays immunosenescence and even rejuvenates immunity in C. elegans. C. elegans gene expression in response to the pathogenic P. aeruginosa strain PA14.

(Submitter supplied) Analysis of differential gene expression in C. elegans adults exposed to three different bacteria: E. coli strain OP50, wild-type P. aeruginosa PA14 and gacA mutant PA14. MAP kinase PMK-1 and transcription factor DAF-16 deletion mutants

Analysis of daf-2(e1368) loss-of-function mutant and daf-2(e1368);pmk-1(km25) and daf-2(e1368);daf-16(mgDf47) double mutants. PMK-1 and DAF-16 confer enhanced pathogen resistance on daf-2 mutants relative to wild-type. Results suggest DAF-16 and PMK-1 upregulate distinct genes to promote immunity.

RNA sequencing analysis of DAF-16 target gene expression when math-33 function is genetically inactivated

(Submitter supplied) In this study we have investigated the effect of loss of math-33 activity on DAF-16-mediated target gene regulation in C. elegans under conditions of reduced Insulin/IGF-1 signaling (IIS). Using whole nematode RNA sequencing experiments we found that the daf-2(e1370)-mediated induction and repression of DAF-16 target genes was decreased in daf-2(e1370); math-33(tm3561) mutant animals. Our data suggest that the downregulation of endogenous DAF-16 isoforms in the absence of a functional MATH-33 severely affects the global expression of DAF-16 targets when IIS activity is reduced. more...

Transcription factor ETS-4 deletion effect on larvae

Analysis of stage L4 larvae with two distinct deletion alleles of ets-4: ets-4(ok165) and ets-4(uz1). Adults with ets-4 mutations have a significant extension of mean life span. Results provide insight into the transcriptional control of aging effectors by the ETS-4 transcription factor.
