Transcriptomic analysis of the porcine anterior pituitary gland during the peri-implantation period

(Submitter supplied) In pigs, the peri-implantation period is recognized as determinant of successful reproduction, and it is controlled by neuroendocrinal interactions among signals originating from the female hypothalamic-pituitary-ovarian axis, uterus, and developing embryo. The transcriptomic activity of the pituitary gland may influence reproductive processes occurring in pigs during the peri-implantation period. The aim of this study was to determine alternations in gene expression in the porcine pituitary gland during the peri-implantation period (days 15-16 of pregnancy) in comparison with that during the respective days of the estrous cycle using a microarray approach.

Transcriptome profile of the anterior pituitary gland in pigs during maternal recognition of pregnancy

(Submitter supplied) The knowledge regarding the reproductive-status-related changes occurring within the transcriptome of the porcine pituitary is limited. This study aimed to compare pituitary gland gene expression profiles of pigs during the maternal recognition of pregnancy period (days 12-13 of pregnancy) with pigs during the respective days of the estrous cycle using a microarray approach. Analysis indicated 482 differentially expressed genes (DEGs) with a FC ≥ 1.5 (p < 0.05) in the pituitary of pregnant vs. estrous-cyclic pigs. more...

Transcriptome Analysis of the Endometrium from Chinese Erhualian sows that differ in Calcium Ion Concentration and Litter Size

(Submitter supplied) Embryonic survival rate is an important factor for the fecundity of sows, and it is affected by endometrium secreting histotroph. A higher concentration of calcium ion has been observed in the uterus of the high prolific Erhualian sows (EH) compared with low prolific ones (EL), which suggests EH sows had a greater establishment and maintenance of pregnancy, and thus increase embryonic survival rate during peri-implantation period. more...

Effect of early calf-hood nutrition on the transcriptomic profile of the HPT axis in Holstein-Friesian bulls

(Submitter supplied) The aim of this study was to investigate the effect of early calf-hood nutrition on the transcriptomic profile of the arcuate nucleus of the hypothalamus, the anterior pituitary and the testes in Holstein-Friesian bulls and possible downstream effects on reproduction. Holstein Friesian bull calves with a mean (±S.D.) age and bodyweight of 19 (±8.2) days and 47.5 (±5.3) kg, respectively, were assigned to either a high (n=10) or low (n=10) plane of nutrition. more...

The in vitro influence of adiponectin on the porcine endometrium on days 15 to 16 of gestation.

(Submitter supplied) Adiponectin is a hormone secreted primarily by white adipose tissue. Recent studies have shown that adiponectin and its receptors (AdipoR1 and AdipoR2) are expressed in different reproductive tissues, including the ovary and uterus. This newly discovered endocrine system plays an important role in the regulation of reproductive processes. The aim of this study was to explore the influence of adiponectin on global gene expression in the porcine endometrium during early gestation, on days 15 to 16 of pregnancy (implantation period).

Comparison between caruncular and intercaruncular areas in the endometrium of cyclic and pregnant cows

(Submitter supplied) At implantation the endometrium undergoes dramatic modifications necessary for its physical interactions with the trophoblast as well as the development of the conceptus. We aim to identify endometrial factors and pathways essential for a successful implantation in the caruncular [C] and the intercaruncular [IC] areas in cattle. The [C versus IC] direct comparison revealed 1177 and 453 DEG in cyclic and pregnant animals respectively, with a major impact of the conceptus on metabolism and cell adhesion. more...

Cycle versus Pregnancy

(Submitter supplied) At implantation the endometrium undergoes dramatic modifications necessary for its physical interactions with the trophoblast as well as the development of the conceptus. We aim to identify endometrial factors and pathways essential for a successful implantation in the caruncular [C] and the intercaruncular [IC] areas in cattle. Using a bovine oligo-array, expression profiles were established at day 20 of the estrous cycle or pregnancy (implantation) showing 446 and 1295 differentially expressed genes (DEG) in [C] and [IC] areas respectively. more...

microRNA expression profile in peri-implantation porcine embryos

(Submitter supplied) Recently, microRNAs (miRNAs) have emerged as new players in the fine tuning of embryo development and implantation in mammals via posttranscriptional gene regulation mechanisms. Applying custom made multispecies arrays we aimed to analyze expression profile of microRNAs in peri-implantation porcine conceptuses/trophoblasts to identify their potential role at the maternal-fetal interface during the critical period of maternal recognition of pregnancy and implantation.

Deep sequencing of the porcine endometrial transcriptome on day 12 of pregnancy

(Submitter supplied) The porcine conceptus undergoes rapid differentiation and expansion of its trophoblastic membranes between Days 11 and 12 of gestation. Concomitant with trophoblast elongation, production of conceptus estrogen, the porcine embryonic pregnancy recognition signal increases. Conceptus attachment to the uterine surface epithelium starts after Day 13 initiating epitheliochorial placentation. To analyze the transcriptome changes in the endometrium in the course of maternal recognition of pregnancy (MRP), deep sequencing of endometrial RNA samples of Day 12 pregnant animals (n=4) and corresponding non-pregnant controls (n=4) was performed using Illumina RNA-Seq. more...

(Submitter supplied) The objective of the present study was to use our own fabricated bovine embryo cDNA microarray to profile genome-wide expression patterns of genes during the implantation period. Duplicate readings were obtained by hybridization of a Cy3/Cy5 probe mixture for each sample. Data normalization was performed the local background intensity of each array spot was smoothed by the local weight regression (lowess) smoother and subtracted from spot intensity data. more...

Transcriptional profiling in the anterior pituitary gland of juvenile broiler chickens genetically selected for high or low body weight

(Submitter supplied) Several processes governing growth and metabolism are regulated by the neuroendocrine system, particularly the trophic hormones produced in the anterior pituitary gland. However, many of the differences that exist in pituitary gene expression and functional gene networks associated with divergent growth characteristics are still largely unknown. Using the Del-Mar 14K Chicken Integrated Systems microarray, this experiment evaluated global gene expression in the anterior pituitary gland during the first 7 weeks of post-hatch growth in two lines of chickens genetically selected for high growth (HG) or low growth (LG) at SRA-INRA, France. more...
