RNA-seq analysis of Control and BSCL2-deleted mature brown adipose tissue in mice

(Submitter supplied) Next generation sequencing was used to compare transcriptome profiling in brown adipose tissue from Ctrl and BKO mice with mature brown adipose tissue specific deletion of BSCL2. Using an optimized data analysis workflow, we identified 13,525 transcripts in brown adipose tissue. 243 genes were differentially expressed with a fold change ≥1.5 and Padj value <0.05. Only 99 genes showed differential expression between the Ctrl and BKO brown fat, with a fold change ≥2.0 and Padj value <0.05. more...

Transcriptional control of thermogenesis by early B cell proteins [ChIP-seq]

(Submitter supplied) Early B Cell Factor 2 (EBF2) is required for brown adipose tissue basal thermogenic gene expression; however, it was unknown whether chronic cold exposure normalized the enhancer landscape of Ebf2 mutant mice. We profiled H3K27Ac binding in Ebf2 WT and mutant brown adipose tissue in mice housed at room temperature or one week of cold exposure.

Single-cell RNA sequencing of primary mouse brown adipocytes

(Submitter supplied) Purpose: To test if there is a heterogeneity within brown adipocytes, we performed single cell RNA-sequencing of primary brown adipocytes isolated from mouse brown adipose tissue. Methods: Brown adipocytes from 10-week-old C57BL/6J male mice were freshly collected and resuspended in PBS containing 0.04% BSA at a concentration of 700~1200 cells/µl. Cell number and viability were measured using a TC20 Automated Cell Counter (BioRad). more...

STAT5 is required for lipid breakdown and beta-adrenergic responsiveness of brown adipose tissue

(Submitter supplied) Increasing energy expenditure through activation of brown adipose tissue (BAT) thermogenesis is an attractive approach to counteract obesity. Thus, it is essential to understand molecular mechanisms that control BAT functions. Here, we describe signal transducer and activator of transcription (STAT) 5 as key regulator of BAT functionality. We found that STAT5 is necessary for acute cold-induced temperature maintenance and stimulated lipid breakdown in BAT using mice that harbour an adipocyte-specific deletion of Stat5a/b genes. more...

(Submitter supplied) Purpose: We investigated the transcriptomic change in brown fat of young and old mice (wild type) through high-throughput RNA-sequencing (RNA-Seq) analysis when the mice were exposed to cold room or room temperatur. Methods: We prepared 10 of young (3 months) mice and 9 of old (24 months) mice, and kept them in cold room (4°c) or room temperature (24°c) for 24 hours. Then, we sacrified mice and extracted RNA from brown fat tissue (BAT) for RNA-seq experiment. more...

Regulation of cold-induced thermogenesis by the stress granule protein FAM195A

(Submitter supplied) Homeothermic vertebrates are capable of surviving in cold environments by maintaining constant body temperature through thermogenesis, in which brown adipose tissue (BAT) produces heat by increasing mitochondrial oxidation along with the uncoupling of the electron transport chain and activation of uncoupling protein 1 (UCP1). Although the transcription factors that control the expression of UCP1 and genes involved in nutrient oxidation, mitochondrial function, have been extensively studied, only a few other proteins essential for BAT function have been identified. more...
