Study of the role of c-Maf in type 2 innate lymphoid cells

(Submitter supplied) Group 2 innate lymphoid cells (ILC2s) are involved in type 2 inflammatory diseases such as allergy. In response to repeated allergen inhalations, lung epithelial cells secrete innate cytokines that activate ILC2s to release interleukin-5 (IL-5) and interleukin-13 (IL-13) resulting in eosinophil infiltration, mucus hyper-production, and airway hyperreactivity. Beside GATA3, very little is known about the role of specific transcription factors in the regulation of ILC2’s function. more...

Gene expression data of naïve and activated ILC2s isolated from female and male mouse lungs

(Submitter supplied) Epidemiological studies have shown sex differences in prevalence of non-allergic asthma. Recent reports demonstrated negative effects of androgen signaling on group 2 innate lymphoid cells (ILC2s), explaining a potential mechanism behind sex bias in asthma prevalence. To further understand the difference in ILC2s based on sex, we have investigated the effects of sex and age on the number and function of lung ILC2s, and epithelium derived cytokines. more...

Expression data from naive, effector and memory-like ILC2s isolated from mouse lungs and those from effector and memory-like ILC2s isolated from mediastinal lymph node

(Submitter supplied) Group 2 innate lymphoid cells (ILC2s) in the lung are stimulated by inhaled allergens. ILC2s do not directly recognize allergens but they are stimulated by cytokines including interleukin (IL)-33 released by damaged epithelium.Lung ILC2s, upon stimulation, produce T helper 2 cell-type cytokines inducing T cell independent allergic lung inflammation. We now report that lung ILC2s, upon activation by an allergen or IL-33, acquire the properties of memory cells. more...

RNA-seq of control and IL-33-treated WT and STAT KO ILC2s in vitro

(Submitter supplied) Group 2 innate lymphoid cell (ILC2), an innate counterpart of T helper 2 cell, plays a critical role in type 2 immune responses. However, the molecular regulation mechanism of ILC2 is still unclear. Here, we find that STAT3 signaling is essential for ILC2 effector function and could drive both acute and chronic allergic inflammation in the lung. Mechanistically, allergic alarmin IL-33 induces a non-canonical STAT3 phosphorylation at serine 727 in ILC2, which leads to its mitochondrial translocation. more...

Next Generation Sequencing Facilitates Quantitative Analysis of the effects of CD200R engagment on sorted mouse pulmonary ILC2 Transcriptomes.

(Submitter supplied) The prevalence of asthma and airway hyperreactivity (AHR) is increasing at an alarming rate. Group 2 innate lymphoid cells (ILC2s) are copious producers of type 2 cytokines, which leads to AHR and lung inflammation. In this study, we demonstrate murine ILC2s express CD200 receptor (CD200R), and this expression is inducible. We ascertain CD200R engagement inhibits activation, proliferation, and type 2 cytokine production, revealing a potent immunoregulatory role for CD200–CD200R axis on ILC2s. more...

MicroRNA regulation of type 2 innate lymphoid cell homeostasis and function in allergic inflammation

(Submitter supplied) MicroRNAs (miRNAs) exert powerful effects on immunity through coordinate regulation of multiple target genes in a wide variety of cells. Group 2 innate lymphoid cells (ILC2s) are tissue sentinel mediators of allergic inflammation. We established the physiological requirements for miRNAs in ILC2 homeostasis and immune function, and compared the global miRNA repertoire of resting and activated ILC2s and T helper type 2 (TH2) cells. more...

Expression profiling of multiple immune cells by high throughput sequencing

(Submitter supplied) ILC2s have the unique ability to produce cytokines such as constitutive production as well as rapid and abundant production during inflammation. We found that the unique type 2 cytokine prodution from ILC2s is regulated by post-transcriptional regulation. Here we identified the genes involved in the post-transcriptional regulation of type 2 cytokine production in ILC2s by comparing the gene expression between multiple immune cells.

Human CD127 negative ILC2s show immunological memory.

(Submitter supplied) Group 2 Innate Lymphoid Cells (ILC2s) are key players in type 2 immunity and contribute to maintaining homeostasis and responding to inflammation. ILC2s are implicated in the development of type 2 inflammation-mediated chronic disorders like asthma. While memory ILC2s have been identified in mouse, it is unknown whether human ILC2s can acquire immunological memory. Here, we demonstrate the persistence of CD45RO, a marker previously linked to inflammatory ILC2s, in resting ILC2s that have undergone prior activation. more...

Bone marrow progenitors and IL-2 signaling contribute to the strain differences of kidney innate lymphoid cells

(Submitter supplied) Innate lymphoid cells (ILCs) are critical mediators of immune responses by their robust production of cytokines. ILCs are mainly present in mucosal tissues, but their regulatory function in the kidney is attracting attention. Nevertheless, the understanding of their unique biology, compared to other tissues, is largely unknown. Here, we aimed to examine the effect of BALB/c and C57BL/6 strains on the number and function of kidney ILCs and their underlying mechanisms. more...

A Diversity of Novel Type-2 Innate Lymphoid Cell Subpopulations Revealed During Tumour Expansion

(Submitter supplied) Type 2 innate lymphoid cells (ILC2s) perform vital functions in orchestrating humoral immune responses, facilitating tissue remodelling, and ensuring tissue homeostasis. Additionally, in a role that has garnered considerably less attention, ILC2s can also enhance Th1-related cytolytic T lymphocyte immune responses against tumours. Studies have thus far generally failed to address the mystery of how one ILC2 cell-type can participate in a multiplicity of functions. more...
