TDP-43, an ALS/FTD causal gene, regulates SREBF2-mediated cholesterol metabolism in the CNS

(Submitter supplied) Maintaining cholesterol homeostasis is essential for health of all animal cells. Because of blood-brain barrier, de novo cholesetrol biosynthesis and intercellular cholesterol transport is thought to maintain cholesterol homeostasis within the central nervous system (CNS). Here, we showed that TDP-43, the pathological signature protein for amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD), regulates SREBF2-mediated cholesterol metabolism in the central nervous system (CNS). more...

GDE2-dependent activation of canonical Wnt signaling in neurons regulates oligodendrocyte maturation

(Submitter supplied) Neurons and oligodendrocytes communicate to regulate oligodendrocyte development and ensure appropriate axonal myelination. Here, we show that Glycerophosphodiester phosphodiesterase 2 (GDE2) encodes a neuronal pathway that promotes oligodendrocyte maturation through the release of soluble neuronally-derived factors. Mice lacking global or neuronal GDE2 expression have reduced mature oligodendrocytes and myelin proteins but retain normal numbers of oligodendrocyte precursor cells (OPCs). more...

SRF transcriptionally regulates the oligodendrocyte cytoskeleton during CNS myelination (snRNA-Seq)

(Submitter supplied) Myelination of neuronal axons is essential for nervous system development. Myelination requires dramatic cytoskeletal dynamics in oligodendrocytes, but how actin is regulated during myelination is poorly understood. We recently identified serum response factor (SRF)—a transcription factor known to regulate expression of actin and actin regulators in other cell types—as a critical driver of myelination in the aged brain. more...

Myelin-allied gene expression profile of adult neural progenitors shortly after onset of differentiation

(Submitter supplied) Spontaneous neural repair from endogenous neural stem cells (NSCs) occurs in response to central nervous system (CNS) injuries or diseases to only a limited extent from endogenous NSCs niches. Uncovering the mechanisms that control neural repair and can be further manipulated to promote towards oligodendrocyte progenitors cells (OPCs) and myelinating oligodendrocytes is a major objective. Our aim was to identify myelin specific transcriptional regulators amongst large transcriptional changes shortly after differentiation of neural stem cells from the subventricular zone (SVZ) of adult mice

Drug-based modulation of endogenous stem cells promotes functional remyelination in vivo

(Submitter supplied) Multiple sclerosis involves an aberrant autoimmune response and progressive failure of remyelination in the central nervous system. Prevention of neural degeneration and subsequent disability requires remyelination through the generation of new oligodendrocytes, but current treatments exclusively target the immune system. Oligodendrocyte progenitor cells are stem cells in the central nervous system and the principal source of myelinating oligodendrocytes. more...

RNA-seq of myelinating oligodendrocytes in Lmna wt and knockout mice

(Submitter supplied) We showed that the composition of the nucler lamina changes with the differentiation state of oligodendrocyte lineage cells, with Lamin B highly expressed in progenitor cells and Lamin A highly expressed in mature oligodendrocytes. The genetic deletion of Lmna in oligodendrocytes results in the onset of a progressive motor phenotype in adult mice. We performed RNA-Seq in myelinating oligodendrocytes expressing the reporter myelin marker NDRG1-EGFP ( Marechal et al., 2021) in the presence or absence of LMNA. more...

TDP-43 represses a cryptic exon in Schwann cells to ensure rapid saltatory conduction

(Submitter supplied) A pathological hallmark of amyotrophic lateral sclerosis and frontotemporal dementia is the aggregation of TDP-43. While extensive exploration into the function of TDP-43 has focused entirely in the central nervous system (CNS), the implications in physiology and pathology of the peripheral nervous system (PNS) have long been overlooked. Herein, we demonstrate that deletion of TDP-43 in Schwann cells results in a 50% reduction in peripheral nerve conduction velocity, without any obvious alterations to peripheral compact myelin. more...

Effect of knockdown of Secisbp2l in primary oligodendrocyte precursor cells

(Submitter supplied) To investigate the function of Secibp2l which we identified as an oligodendrocyte-specific RNA binding protein, we performced transcriptome profiling of primary oligodnedrocytes with Secisbp2l knockdown by siRNA. Cytotoxic CNS-associated T cells drive axon degeneration in aging and myelin disease

(Submitter supplied) 1. Aging is a major risk factor for the development of nervous system functional decline, even in the absence of diseases or trauma. The axon–myelin units and synaptic terminals are some of the neural structures most vulnerable to aging-related deterioration, but the underlying mechanisms are poorly understood. In the peripheral nervous system, macrophages—important representatives of the innate immune system—are prominent drivers of structural and functional decline of myelinated fibers and motor endplates during aging. more...
