Long-read transcriptome sequencing reveals expression characteristics of osteosarcoma

(Submitter supplied) Osteosarcoma is the most common primary bone cancer in children, adolescents and young adults. It is a rare cancer type. To comprehensively reveal the transcriptomic characteristics of osteosarcoma, we performed Oxford Nanopore Technologies (ONT) long-read RNA-Seq of tumor and adjacent normal tissues from 23 patients with osteosarcoma.

RNA Sequencing Quantitative Analysis of cell lines derived from tumours from two mouse models of human osteosarcoma that recapitulate clinically distinct human osteosarcoma subtypes.

(Submitter supplied) Purpose: Osteosarcoma (OS) is the most common primary bone malignancy. OS consists of several subtypes including fibroblastic, osteoblastic and chondroblastic OS. We have developed genetically engineered mouse models of human OS that recapitulate two distinct subtypes, fibroblastic (Osx-CreLox p53-/- Rb-/-) and osteoblastic (Osx-Cre shRNA p53-/-) OS. The goal of this study was to identify transcriptional differences that distinguish the two subtypes. more...

RNA sequencing of Cd44-positive and Cd44-negative primary osteosarcoma isolated from Nf2 - mutant mice

(Submitter supplied) Osteosarcoma is the most common type of pediatric bone tumor. Despite great advances in chem-otherapy during the past decades the survival rates of osteosarcoma patients remains usatisfacto-ry. Drug resistance is one of the main reasons leading to treatment failure and poor progno-sis. Previous reports correlated expression of cluster of differentiation 44 (CD44) with drug re-sistance and poor survival of osteosarcoma patients, however the underlying mechanisms are poorly defined. more...

Identification of osteosarcoma driver genes by integrative analysis of copy number and gene expression data

(Submitter supplied) High-grade osteosarcoma is a tumor with a complex genomic profile, occurring primarily in adolescents with a second peak at middle age. The extensive genomic alterations obscure the identification of genes driving tumorigenesis during osteosarcoma development. In order to identify such driver genes, we integrated DNA copy number profiles (Affymetrix SNP 6.0) of 32 diagnostic biopsies with 84 expression profiles (Illumina Human-6 v2.0) of high-grade osteosarcoma as compared with its putative progenitor cells, i.e. more...

Genome-wide gene expression analysis of high-grade osteosarcoma

(Submitter supplied) We performed genome-wide gene expression data of high-grade osteosarcoma samples. In order to detect osteosarcoma drivers, we integrated these data with copy number data. We performed two different methods - a non-paired and a paired integrative analysis.
