ERK MAP kinase inactivation during G0/G1 to S phase transition: time course

Analysis of NIH3T3 cells treated with the ERK inhibitor U0126 before or after the addition of the ERK activator FGF. Cells stimulated with FGF for up to 12 hours. Results identify genes regulated by the ERK pathway during G0/G1 to S phase transition.

RNA mapping and ChIP with tiling arrays in FGF-stimulated NIH3T3 cells

(Submitter supplied) Transcription initiation in mammals is assumed to be independently controlled among nearby genes. On the other hand, recent large-scale transcriptome analyses have shown that the genome is pervasively transcribed, such that the majority of its DNA gives rise to RNAs. A simple question therefore arises: is it possible to pinpoint and activate a particular locus without perturbing numerous neighboring transcripts? Here we show that intensive transcription of immediate-early genes (IEGs) in response to growth factor stimulation is often accompanied by co-upregulation of their neighboring genes. more...

Temporal control of gene expression by ERK MAP kinase during cell cycle progression from G0/G1 to S phase

(Submitter supplied) The ERK family of MAP kinase plays a critical role in growth factor-stimulated cell cycle progression from G0/G1 to S phase. But, how sustained activation of ERK promotes G1 progression has remained unclear. Here, our systematic analysis on the temporal program of ERK-dependent gene expression shows that sustained activation of ERK is required for induction and maintenance of the decreased expression levels of a set of genes. more...

Requirement for ERK MAP kinase in mouse preimplantation development

(Submitter supplied) Preimplantation development is a crucial step for successful implantation and pregnancy. Although both compaction and blastocyst formation have been extensively studied, mechanisms regulating early cell division stages before compaction have remained unclear. Here, we show that ERK MAP kinase function is required for early embryonic cell division and normal cell-cell adhesion before compaction. Our analysis demonstrates that inhibition of ERK activation in the late 2-cell stage embryos leads to a reversible arrest in G2 phase in the 4-cell stage. more...

MicroRNA expression during cell cycle arrest

(Submitter supplied) The miR-16 family, which targets genes important for the G1-S transition, is a known modulator of the cell cycle, and members of this family are often deleted or down-regulated in many types of cancers. Here we report the reciprocal relationship - that of the cell cycle controlling the miR-16 family. Levels of this family increase rapidly as cells are arrested in G0. Conversely, as cells are released from G0 arrest, levels of the miR-16 family rapidly decrease. more...

Hela Mitotic Selection - gene expression time series in early G1 phase of the human cell cycle

(Submitter supplied) HeLa mitotic cells were collected using a mitotic shakeoff apparatus to study gene expression in early G1 phase of the human cell cycle. This set includes microarray data from two shake-off experiments : shake 1 (0h-14h) which was conducted over 14 hours and RNA samples collected every 2 hrs, and shake 2 (0min-120min) which was conducted over 2 hours and samples collected every 15 minutes. Total RNA was prepared using ULTRASPEC RNA isolation system, and Reference RNA was extracted from asynchronously growing HeLa cells using TRIzol. more...
