Bis(2-chloroethoxy)methane effect on hearts: time course

Analysis of hearts exposed to the heart toxin bis(2-chloroethoxy)methane (CEM) for 2 or 5 days. Toxicity occurs at day 2 of CEM dosing, and recovery at day 5 despite continued dosing. Results provide insight into the molecular basis of heart toxicity and recovery during the exposure to CEM.

Bis(2-chloroethoxy)methane-induced heart toxicity and recovery in male fisher rats

(Submitter supplied) Specific gene changes after exposure to the heart toxin, bis(2-chloroethoxy)methane, were analyzed to elucidate critical pathways associated with recovery from heart toxicity. Bis(2-chloroethoxy)methane was administered to male F344/N rats at 0, 200, 400, or 600 mg/kg by dermal exposure for up to 16 days. Heart toxicity occurred at day 2 of exposure in all three regions of the heart (atrium, ventricle, interventricular septum) and was characterized by myofiber vacuolation, necrosis, mononuclear-cell infiltration, and atrial thrombosis. more...

Sperm mRNA transcripts are indicators of sub-chronic low dose testicular injury in the Fischer 344 rat

(Submitter supplied) Current human reproductive risk assessment methods rely on semen and serum hormone analyses, which are not easily comparable to the histopathological endpoints and mating studies used in animal testing. Because of these limitations, there is a need to develop universal evaluations that reliably reflect male reproductive function. We hypothesized that toxicant-induced testicular injury can be detected in sperm using mRNA transcripts as indicators of insult. more...

Analysis of Akt1 activation in transgenic mouse hearts show expression profiles associated with hypertrophy and failure

(Submitter supplied) To investigate molecular mechanisms involved in the development of cardiac hypertrophy and heart failure, a tetracycline-regulated transgenic system to conditionally switch a constitutively-active form of the Akt1 protein kinase on or off in the adult heart was developed. Short-term activation (2 weeks) of Akt1 resulted in completely reversible hypertrophy with maintained contractility. In contrast, chronic Akt1 activation (6 weeks) induced extensive cardiac hypertrophy, severe contractile dysfunction, and massive interstitial fibrosis. more...

Expression data from brain tissue of Rattus norvegicus treated with D-Serine

(Submitter supplied) d-serine is naturally present throughout the human body. It is also used as add-on therapy for treatment-refractory schizophrenia. d-Serine interacts with the strychnine-insensitive glycine binding site of NMDA receptor, and this interaction could lead to potentially toxic activity (i.e., excitotoxicity) in brain tissue. The transcriptomic changes that occur in the brain after d-serine exposure have not been fully explored. more...

D-serine effect on the brain: dose response

Analysis of forebrains of animals treated with up to 500 mg/kg D-serine for 96 hours. D-serine is involved in many physiological processes through its interaction with the glycine binding site of the NMDA receptor. Results provide insight into the impact of D-serine exposure on neuronal functions.

Aging-Induced Alterations in Gene Transcripts and Functional Activity of Mitochondrial Oxidative Phosphorylation Complexes in the Heart

(Submitter supplied) Aging is associated with progressive decline in energetic reserves compromising cardiac performance and tolerance to injury. Although deviations in mitochondrial functions have been documented in senescent heart, the molecular bases for the decline in energy metabolism are only partially understood. High-throughput transcription profiles of genes coding for mitochondrial proteins in ventricles from adult and aged rats were compared using microarray analysis.

Short-term growth hormone treatment on the liver and muscle transcriptome in rainbow trout (Oncorhynchus mykiss)

(Submitter supplied) Although studies have established that exogenous growth hormone (GH) treatment stimulates growth in fish, its effects on target tissue gene expression are not well characterized. We assessed the effects Posilac® (Monsanto Co., St. Louis, MO), a recombinant bovine somatotropin, on tissue transcript levels. Transcript abundance was measure in liver and muscle using the GRASP 16 K cDNA microarray. A selection of the genes identified as altered with the microarray, and also transcripts for insulin-like growth factors, growth hormone receptors (GHR) and myostatins were measured by realtime PCR in the liver, muscle, brain, kidney, intestine, stomach, gill and heart. more...

Transcriptional Response to Aging and Caloric Restriction in Heart and Adipose Tissue

(Submitter supplied) Sustained caloric restriction (CR) extends lifespan in animal models but the mechanism and primary tissue target(s) have not been identified. Gene expression changes with aging and CR were examined in both heart and subcutaneous white adipose tissue (WAT) of F344 male rats using Affymetrix® RAE 230 arrays and validated by qRT-PCR on 18 genes. In heart, age- associated changes but not CR-associated changes in old. more...

Mitochondrial functional aberrations in a mouse model of non-ischemic heart failure

(Submitter supplied) We characterized the metabolic and cardiac mitochondrial function in a mouse model of non-ischemic HF. Inhibition of nitric oxide synthesis and hypertension, which often present together, are two important risk factors in human non-ischemic HF. Compared with L-NAME L-NG-Nitroarginine methyl ester (L-NAME), an inhibitor of nitric oxide synthesis or Angiotensin II (AngII), a hypertensive agent treatment alone, L-NAME+AngII induced the most severe HF phenotype characterized by edema, hypertrophy, fibrosis, increased blood pressure and reduced ejection fractions. more...

Identification of a responsive gene set to evaluate the potential impact of seismic exposure on Atlantic salmon (Salmo salar) inner ear

(Submitter supplied) Considerable interest and controversy has arisen over the potential effects of seismic surveys carried out during exploration for oil and gas deposits. Regarding fish, there is a concern that intense sound sources, such as seismic airguns, may injure their auditory system. In this study, salmonid cDNA microarrays and quantitative reverse transcription – polymerase chain reaction (QPCR) were used to identify and study a responsive gene set in the inner ear of Atlantic salmon (Salmo salar) following seismic airgun exposure. more...
