Pulmonary microvascular endothelial cell response to hypoxia in vitro: time course

Analysis of pulmonary microvascular endothelial cells subjected to hypoxia for up to 48 hours. Pulmonary hypoxia is a common complication of chronic lung diseases leading to the development of pulmonary hypertension. Results provide insight into the molecular pathogenesis of pulmonary hypertension.

Lung selective gene responses to alveolar hypoxia

(Submitter supplied) Pulmonary hypoxia is a common complication of chronic lung diseases leading to the development of pulmonary hypertension. The underlying sustained increase in vascular resistance in hypoxia is a response unique to the lung. Thus, we hypothesised that there are genes whose expression is altered selectively in the lung in response to alveolar hypoxia. Using a novel subtractive array strategy, we compared gene responses to hypoxia in primary human pulmonary microvascular endothelial cells to those in cardiac microvascular endothelium and identified genes selectively differentially regulated in the lung endothelium. more...

Tumor and Non Tumor Associated Skin Fibroblasts

(Submitter supplied) Fibroblast cells were cultured from human skin, either tumor (basal cell carcinoma) or non-tumor tissue. Cells were propagated for several passages in culture, then switched to low (0.1%) serum conditions for 48 hours prior to mRNA harvest. Reference for all arrays was Stratagene Universal Human Reference RNA. Set of arrays organized by shared biological context, such as organism, tumors types, processes, etc. more...

Basal Cell Carcinoma Tumor-derived Fibroblasts

(Submitter supplied) Dataset contains samples that are all fibroblast cells that have been cultured from fresh human tissue. Fibroblasts were derived either from tumor (basal cell carcinoma) or non-tumor (control) tissue. Samples were maintained in DMEM, 10% FBS, glutamine, pen/strep and passaged for 4 passages before RNA harvest was performed using Invitrogen FastTrack. Reference channel RNA is the same for each array in the dataset and is Stratagene Universal Reference RNA. more...

Estrogen receptor-dependent attenuation of hypoxia-induced changes in the lung genome of pulmonary hypertension rats

(Submitter supplied) 17β-estradiol (E2) exerts complex and context-dependent effects in pulmonary hypertension. In hypoxia-induced pulmonary hypertension (HPH), E2 attenuates lung vascular remodeling through estrogen receptor (ER)-dependent effects; however, ER target genes in the hypoxic lung remain unknown. In order to identify the genome regulated by the E2-ER axis in the hypoxic lung, we performed a microarray analysis in lungs from HPH rats treated with E2 (75 mcg/kg/d) ± ER-antagonist ICI182,780 (3 mg/kg/d). more...

Transcriptomics revealed sex differences in human pulmonary microvascular endothelial cells in normoxia and hypoxia

(Submitter supplied) To investigate sex differences at the transcriptome level in human pulmonary microvascular endothelial cells (HPMECs) from healthy male and female donors basally (in normoxia) and in hypoxic conditions. RNA-seq was performed on male (n=3) and female (n=4) HPMECs that were cultured in conditions of physiological shear stress (PMID: 36730645) in normoxia (21% O2) or in hypoxia (1% O2) for either 24 or 48 hours.
