Analysis of differentiating E14 TG2a ESCs with cell surface expression of 5T4. The upregulation of 5T4 expression is associated with loss of pluripotency in ES cells. Results provide insight into molecular mechanisms underlying the balance between pluripotency and differentiation in ES cells.

Dissection of human embryonic stem cells-derived endoderm progenitors by differential cell capture on antibody arrays

(Submitter supplied) Heterogeneity, shortage of material, and lack of progenitor-specific cell surface markers are major obstacles to elucidating the mechanisms underlying developmental processes. Here we report a proteomic platform that alleviates these difficulties and demonstrate its effectiveness in fractionating heterogeneous cultures of early endoderm derived from human embryonic stem cells. RNA-seq analysis of differentially expressed genes as a function of initial cell-population density during lineage-commitment in mouse embryonic stem cells

(Submitter supplied) We examined all transcriptome-level expressions in three initial cell-population densities (862, 1724 and 5172 cells/cm2) in the first two days of differentiation in N2B27. We collected cells in 10-mL tubes and centrifuged them using a pre-cooled centrifuge. We then extracted RNA from each cell-pellet using the PureLink RNA Mini Kit (Ambion, Life Technologies) according to its protocol.
