Tibial muscular dystrophy

Analysis of muscles of tibial muscular dystrophy (TMD) patients. TMD is a late onset, autosomal dominant distal myopathy of the anterior lower leg that results from titin mutations. Results provide insight into the molecular events leading from the causative mutations to the death of muscle cells.

Gene expression profiling in tibial muscular dystrophy and control skeletal muscle

(Submitter supplied) Tibial muscular dystrophy (TMD) is a late onset, autosomal dominant distal myopathy that results from mutations in the two last domains of titin. The cascade of molecular events leading from the causative Titin mutations to the preterm death of muscle cells in TMD is largely unknown. To identify these components, we used gene expression profiling of muscle biopsies from TMD patients and healthy controls.

Real-time quantitative PCR analysis of UPR genes in human normal and osteoarthritic chondrocytes

(Submitter supplied) Human chondrocytes were obtained from the cartilage of femoral condyles and tibial plateaus from individuals within 12 hours of death (normal) and from patients undergoing total knee arthroplasty (osteoarthritic, OA). The chondrocytes were released from the cartilage, seeded at high density, and cultured in Dulbecco’s modified Eagle’s medium supplemented with 10% heat-inactivated fetal bovine serum and an antibiotic mixture (100 units/ml penicillin base and 100 μg/ml streptomycin base) at 37°C in a humidified atmosphere of 5% CO2/95% air. more...

Transcriptome of Arabidopsis thaliana ceh1 mutant

(Submitter supplied) Here we present the transcriptomic profile of mutant plants designated as ceh1 (constitutively expressing HPL). CEH1 encodes 1-hydroxy-2-methyl-2-butenyl 4-diphosphate synthase (HDS), the enzyme controlling the bottleneck step of the biosynthesis of isopentenyl diphosphate via the 2-C-methyl-d-erythritol-4-phosphate (MEP) pathway in the plastids. Mutation of this enzyme in ceh1 mutant led to accumulation of high levels of the stress specific signaling metabolite 2c-methyl-D-erythritol 2,4-cylclodiphosphate (MEcPP), and consequently constitutive activation of a selected otherwise stress responsive genes. more...
