Transcription factor EB overexpression effect on brain hippocampus with an accumulation of mutant tau deposits

Analysis of hippocampi of Tg4510 animals overexpressing transcription factor EB (TFEB). Tg4510 animals, a model for Alzheimer’s disease (AD), express the P301L mutant microtubule-associated protein tau. Results provide insight into the molecular effects of TFEB on tauopathy diseases including AD.

TFEB-vacuolar ATPase signaling regulates lysosomal activity and immune response in tauopathy

(Submitter supplied) Transcription factor EB (TFEB) mediates gene expression through binding to the Coordinated Lysosome Expression And Regulation (CLEAR) sequence. We created a knock-in mouse line in which the CLEAR sequence of ATP6V1H was mutated. Crossing the CLEAR mutant with PS19 mice led to higher tau pathology but dampened glial cell activation. snRNA-seq revealed that the CLEAR mutant failed to be activated in tauopathy conditions.

Since RBP aggregation may result in a loss of normal RBP function, we examined whether dysregulation of RNA metabolism contributes to tauopathies.
