Early postnatal skeletal muscle stem cells: time course

Analysis of myogenic stem cells called satellite cells (mSCs) FACS-sorted from the trunk of Pax3GFP/+ mice at postnatal days 1, 12, and 28, a period when most mSCs are in proliferation and quiescence states. Results provide insight into molecular basis of early postnatal skeletal muscle development.

Early postnatal expression data from mouse skeletal muscle stem cells

(Submitter supplied) Satellite cells are the primary source of stem cells for skeletal muscle growth and regeneration. Since adult stem cell maintenance involves a fine balance between intrinsic and extrinsic mechanisms, we performed genome-wide chronological expression profiling to identify the transcriptomic changes involved during early postnatal growth till acquisition of satellite cell quiescence.

Induction of muscle stem cell quiescence by the secreted niche factor Oncostatin M

(Submitter supplied) Regeneration of skeletal muscle is dependent on the function of tissue-resident muscle stem cells (MuSC), known as satellite cells. MuSC dysfunction is central to muscle pathophysiology, including in age-associated loss of muscle regenerative capacity and congenital disorders such as Duchenne muscular dystrophy. Despite the central role of satellite cells in muscle regeneration, the signals controlling the balance between muscle stem cell quiescence, proliferation, and differentiation remain incompletely understood. more...

Notch signaling/MyoD double deficient embryonic muscle progenitors

(Submitter supplied) Skeletal muscle growth and regeneration rely on myogenic progenitor and satellite cells, the stem cells of postnatal muscle. Elimination of Notch signals during mouse development results in premature differentiation of myogenic progenitors and formation of very small muscle groups. Here we show that this drastic effect is rescued by mutation of the muscle differentiation factor MyoD. However, rescued myogenic progenitors do not assume a satellite cell position and contribute poorly to myofiber growth. more...

Genome wide expression analysis of FACS-isolated Shp2 mutant neonatal myogenic progenitor cells

(Submitter supplied) The equilibrium between proliferation and quiescence of myogenic progenitor and stem cells is tightly regulated to ensure appropriate muscle growth and repair. The non-receptor tyrosine phosphatase Shp2 (Ptpn11) is an important transducer of growth factor and cytokine signaling. Here we combined complex genetic analyses, biochemical studies and pharmacological interference to demonstrate a central role of Shp2 in postnatal myogenesis of mice. more...

Gene profiling of quiescent and activated skeletal muscle satellite cells by an in vivo approach

(Submitter supplied) The satellite cell of skeletal muscle provides a paradigm for quiescent and activated tissue stem cell states. We have carried out transcriptome analyses by comparing satellite cells from adult skeletal muscles, where they are mainly quiescent, with cells from growing muscles, regenerating (mdx) muscles, or with cells in culture, where they are activated. Our study gives new insights into the satellite cell biology during activation and in respect with its niche. more...

RNA-seq profiling of C/EBPβ-overexpressing myoblasts [proliferating myoblasts]

(Submitter supplied) We previously identified C/EBPβ as an inhibitor of myogenic differentiation and a regulator of muscle satellite cell self-renewal. We found that C/EBPβ is regulated during the transition from proliferation to growth arrest in myoblasts and overexpression of C/EBPβ in myoblasts promotes cell growth arrest in vitro and improves engraftment of satellite cells into the stem cell niche in vivo. To identify the molecular mechanism by which C/EBPβ regulates myoblast proliferation and growth arrest, we performed RNA-seq on C/EBPβ-overexpressing myoblasts.

Combination of high-dimensional approaches for skeletal muscle tissue cartography

(Submitter supplied) The regenerative capacity of skeletal muscle relies on muscle stem cells (MuSCs, or satellite cells) and its niche interactions with different neighboring cells. To understand the cellular diversity within adult skeletal muscle tissue, we harvested mononuclear cells from hindlimb skeletal muscles, sorted into single cells and profiled them by single-cell RNA-seq. To further understand and compare the expression profile between MuSCs and a novel smooth-muscle/mesenchymal-like cells (SMMCs) population, we isolated the two cell types by FACS and profiled them respectively by bulk RNA-seq.

Dynamic binding of RBPJ is determined by Notch signalling status

(Submitter supplied) Notch signalling plays crucial roles in mediating cell fate choices in all metazoans largely by specifying the transcriptional output of one cell in response to a neighbouring cell. The DNA-binding protein RBPJ is the principle effector of this pathway in mammals and together with the transcription factor moiety of Notch (NICD) it regulates the expression of target genes. The prevalent view presumes that RBPJ statically occupies consensus binding sites while exchanging repressors for activators in response to NICD. more...

Gene Expression Changes in Soleus Muscle in Mice after Pericytes-Deletion

(Submitter supplied) Pericytes(PCs) are mural cells embedded in the capillary basal lamina. Some PC populations exhibit multipotency, similar to mesenchymal stem cells. Here, the the process of adaptation was investigated by determining changes in transcript profiles when NG2 positive PCs were deleted. PC-deletion was induced by using NG2CreER(+/-)/diphtheria toxin flagment A(DTA) transgenic mice. Cre recombinase was induced by tamoxifen(Tam) treatment(intraperitoneally, 100mg/kg body weight per day) for 5days. more...

Comparison of Wild type and Pofut1-deleted skeletal muscle

(Submitter supplied) Pofut1 is an essential gene that glycosylates proteins containing EGF-like repeats, including Notch Receptors (NotchRs).  Work in mice and in Drosophila has shown that O-fucosylation by Pofut1 is required for NotchR ligands to bind to and activate NotchRs.  As such, Pofut1 deletion in skeletal myofibers allows for an analysis of potential functions and molecular changes of Pofut1 in skeletal muscle that derive from its expression in skeletal myofibers. more...

Increased activity of canonical NF-kB signaling in muscle fibers alters the satellite cell niche and restrains muscle stem cell function during aging

(Submitter supplied) It has been known for some time that muscle repair potential becomes increasingly compromised with advancing age, and that this age-related defect is associated with reduced activity of muscle satellite cells and with the presence of chronic, low grade inflammation in the muscle. Working from the hypothesis that a heightened inflammatory tone in aged muscle could contribute to poor regenerative capacity, we developed genetic systems to inducibly alter inflammatory gene expression in satellite cells or muscle fibers by modulation of the activity of nuclear factor κB (NF-κB), a master transcriptional regulator of inflammation whose activity is upregulated in many cell types and tissues with age. more...

Identification of underexplored mesenchymal and vascular-related cell populations in human skeletal muscle

(Submitter supplied) Skeletal muscle repair and maintenance are directly and indirectly supported by stromal populations such as vascular cells and fibro-adiopgenic progenitors (FAPs), a subset of which express Twist2 and possess direct myogenic potential in rodents. However, less is understood of the complexity and heterogeneity of human skeletal muscle stromal cells. To this end, we performed single-cell RNA-sequencing on ~2000 cells isolated from the human semitendinosus muscle of young individuals. more...
