Hormone-induced adipogenesis suppressed by 2,3,7,8-tetrachlorodibenzo-p-dioxin and EGF

Analysis of differentiated C3H10T1/2 fibroblasts treated with 10 nM 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and/or 10 nM EGF. Prior to treatment, cells induced to differentiate into adipocytes with IDMB hormone. EGF increases effectiveness of TCDD in suppressing hormone-induced adipogenesis.

C3H10T1/2 Adipogenesis: Effects of EGF and TCDD

(Submitter supplied) Confluent C3H10T1/2 cells stimulated to differentiate using IDMB (1uM BRL49653, 1uM Dexamethasone, 0.5uM IBMX, 10ug/mL Insulin). Fetal mouse heart, TCDD dose-response series

(Submitter supplied) Pregnant C57Bl6N mice were treated with 0 (corn oil), 1.5, 3.0, or 6.0 ug/kg TCDD on gd14.5. Fetal hearts were collected on gd17.5. Hearts from each litter were pooled onto one chip. 4 replicates of each condition were run on affymetrix MG_U74Av2 chips, using standard affymetrix protocols and controls. Analysis of gestational day (GD) 17.5 fetal hearts isolated from pregnant C57BL/6Ns treated at GD 14.5 with up to 6.0 ug/kg 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). TCDD, a cardiovascular teratogen, inhibits cardiomyocyte proliferation, a process that peaks between GD 14.5 to 17.5.

Transcriptional Profiling of rat hypothalamic response to TCDD

(Submitter supplied) In many mammals, halogenated aromatic hydrocarbon (HAH) exposure causes wasting syndrome, defined as lethal weight loss as a result of severe and persistent hypophagia. The most potent HAH in causing wasting is 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD), which exerts its toxic effects through the aryl hydrocarbon receptor (AHR) – a transcription factor. Inter-strain Heterogeneity in Responses to TCDD

(Submitter supplied) We profiled hepatic transcriptional responses of 6 strains of rats with varying sensitivity to a dioxin, TCDD, at 19 hours following exposure. The resistant rats exhibited significantly reduced transcriptional responses in comparison to the sensitive strains. We hypothesize that genes which show differential changes between the resistant and sensitive rats may potentially explain sensitivity.

TCDD effect on regenerating fin: time course
