Single cell ATAC sequencing profile of quiescent mesenchymal progenitors

(Submitter supplied) After tissue injury mesenchymal progenitor subsets undergo a transient proliferative expansion, morphological transition and migration from their perivascular niche. Some of these subsets contribute enduring populations to connective tissue structures adjacent to muscle. To gain insights into gene accessibility to complement the single cell RNA seq expression data previously obtained and to uncover the lineage potential of MPs in this context, tdTomato positive cells were profiled by single cell ATAC sequencing.

RNA-seq profiling of adult skeletal muscle after long term conditional knockout of Hic1 [TA RNA-seq]

(Submitter supplied) To determine the effects of long term deletion of the transcriptional repressor Hypermethylated in cancer 1 (Hic1) on skeletal muscle, samples were collected of both wild type mice and those conditionally deleted for Hic1 and profiled by RNA sequencing. Aspects of muscle homeostasis were negatively impacted, highlighting cell and non cell autonomous roles for Hic1 in maintaining functional MPs and muscle tissue.

To gain transcriptomic insights into the MP function in regeneration, a muscle-injury model was employed to activate MPs and follow their contribution to tissue regeneration. To compliment the population RNA seq profile previously obtained and to uncover the suspected heterogeneity of MPs in this context, tdTomato positive cells from selected timepoints were profiled by single cell RNA sequencing.

To gain transcriptomic insights into the transition of MPs out of the quiescent state and their role in muscle regeneration, a muscle-injury model was used to activate MPs and follow their contribution to tissue regeneration. tdTomato positive MPs were FACS purified from dissociated muscle at different time points after notexin-induced injury and transcriptomic analysis was performed using RNA sequencing of all samples.

RNA-seq profiling of FACS purified MPs from adult skeletal muscle after conditional knockout of Hic1 [MP RNA-seq]

(Submitter supplied) To determine the effect of the deletion of the trancriptional repressor Hypermethylated in cancer 1 (Hic1) on quiescence, adult mesenchymal progenitors (MPs) were isolated from skeletal muscle of both wild type mice and those conditionally deleted for Hic1 and profiled by RNA sequencing. Comparison to MPs activated by using a muscle injury model of the tibialis anterior muscle revealed similar pathways affected under both conditions, highlighting the role for Hic1 in maintaining the quiescent state of MPs
