The Parasite Intraerythrocytic Cycle and Human Circadian Cycle are Coupled During Malaria Infection

(Submitter supplied) During infections with malaria parasites P. vivax, patients exhibit rhythmic fevers every 48 hours.  These fever cycles correspond with the time parasites take to traverse the Intraerythrocytic Cycle (IEC) and may be guided by a parasite-intrinsic clock.  Different species of Plasmodia have cycle times that are multiples of 24 hours, suggesting they may be coordinated with the host circadian clock. We utilized an ex vivo culture of whole blood from patients infected with P. vivax to examine the dynamics of the host circadian transcriptome and the parasite IEC transcriptome.  Transcriptome dynamics revealed that the phases of the host circadian cycle and the parasite IEC were correlated across multiple patients, suggesting that the cycles are coupled.  In mouse model systems, host-parasite cycle coupling appears to provide a selective advantage for the parasite.  Thus, understanding how host and parasite cycles are coupled in humans could enable anti-malarial therapies that disrupt this coupling.

Global gene expression of human malaria parasite liver stages throughout intrahepatocytic development

(Submitter supplied) To investigate the poorly characterized asymptomatic liver stages (LS) of plasmodium faciparum, we analyzed isoloated LS RNA which is the first comprehensive gene expression profile during in vivo intrahepatocytic development. We identified LS-specific processes such as cell cycle, metabolism, and host-parasite interaction pathways, and, interestingly, var gene transcipts.

A single-cell liver atlas of relapsing human malaria [Bulk-seq]

(Submitter supplied) Malaria-causing Plasmodium vivax parasites can linger in the human liver for weeks to years, and then reactivate to cause recurrent blood-stage infection. While an important target for malaria eradication, little is known about the molecular features of the replicative and non-replicative states of intracellular P. vivax parasites, or their human host-cell dependencies and the host responses to them. more...

Transcriptome profiling of Plasmodium vivax in Saimiri monkeys identifies potential ligands for invasion

(Submitter supplied) Unlike in Asia and Latin America, Plasmodium vivax infections were rare in Sub-Saharan Africa due to the absence of the Duffy blood group antigen (Duffy Antigen), the only known erythrocyte receptor for the P. vivax merozoite invasion ligand, Duffy Binding Protein 1 (DBP1). However, P. vivax infections have been documented in Duffy-negative individuals throughout Africa, suggesting that P. vivax may use ligands other than DBP1 to invade Duffy-negative erythrocytes through other receptors. more...

Unraveling the Plasmodium vivax sporozoite transcriptional journey from mosquito vector to human host

(Submitter supplied) Malaria parasites transmitted by mosquito bite are remarkably efficient in establishing human infections. The infection process requires ~30 minutes and is highly complex as quiescent sporozoites injected with mosquito saliva must be rapidly activated in the skin, migrate through the body, and infect the liver. This process is poorly understood for Plasmodium vivax due to low infectivity in the in vitro models. more...

RNAseq of hypnozoites and schizonts in the MPCC system

(Submitter supplied) Upon P. vivax infection, dormant liver stage forms, hypnozoites, linger for weeks to months and then relapse to cause recurrent blood stage infection. Very little is known about the hypnozoite: Definitive biomarkers are lacking and in vitro platforms that support phenotypic studies have not been available. Here, we recapitulate the entire liver stage life cycle of P. vivax in vitro in a multiwell format and assay the hypnozoite transcriptome

Comparative profiling of the heterochromatin landscape in different life cycle stages, strains and species of malaria parasites

(Submitter supplied) Heterochromatin is a tightly packaged form of DNA that leads to permanent or temporal gene silencing. In P. falciparum heterochromatin is formed after trimethylation of lysine 9 on histone H3 and consequent binding of heterochromatin protein 1 (HP1). Genome-wide profiling studies established that in P. falciparum blood stage schizonts heterochromatin is formed at subtelomeres and some intra-chromosomal islands. more...

A custom genome-wide Plasmodium vivax 8x15K microarray for expression profiling of Indian clinical isolates showing complicated malaria

(Submitter supplied) High density oligonucleotide microarrays have been used on Plasmodium vivax field isolates to estimate whole genome expression. However, no microarray platform has been experimentally optimized for studying the transcriptome of field isolates. In the present study, we adopted both bioinformatics and experimental testing approaches to select best optimized probes suitable for detecting parasite transcripts from field samples and included them in designing a custom 15K P. more...

Common asymptomatic and submicroscopic malaria infections in Western Thailand revealed in longitudinal molecular and serological studies: a challenge to malaria elimination.

(Submitter supplied) Malaria is the most important vector-borne disease in Southeast Asia. In Thailand, malaria incidence has been in decline, with the annual parasite incidence dropping to 0.56 in 2007. The Myanmar-Thai border province of Tak is considered meso-endemic for malaria, and both Plasmodium vivax (Pv) and P. falciparum (Pf) are equally present. As part of the International Centers for Excellence in Malaria Research (ICEMR) - Southeast Asia project, malaria surveillance is conducted in Tak on both the healthy population and hospital patients, and parasite prevalence is reportedly <1%. more...

Submicroscopic and asymptomatic Plasmodium falciparum and P. vivax infections are common in western Thailand - molecular and serological evidence

(Submitter supplied) Background: Malaria is a public health problem in parts of Thailand, where Plasmodium falciparum and Plasmodium vivax are the main causes of infection. In the northwestern border province of Tak parasite prevalence is now estimated to be less than 1% by microscopy. Nonetheless, microscopy is insensitive at low-level parasitaemia. The objective of this study was to assess the current epidemiology of falciparum and vivax malaria in Tak using molecular methods to detect exposure to and infection with parasites; in particular, the prevalence of asymptomatic infections and infections with submicroscopic parasite levels. more...

Natural anitsense transcripts in Plasmodium vivax clinical isolates

(Submitter supplied) Plasmodium vivax is the most geographically widespread human malaria parasite causing approximately 130-435 million infections annually. It is an economic burden in many parts of the world and poses a public health challenge along with the other Plasmodium sp. The biology of this parasite is very little understood. Emerging evidences of severe complications due to infections by this parasite provides an impetus to focus research on the same. more...

Whole-genome sequencing and microarray analysis of ex vivo Plasmodium vivax reveal selective pressure on putative drug resistance genes

(Submitter supplied) Plasmodium vivax causes 25-40% of malaria cases worldwide, yet research on this human malaria parasite has been neglected. Nevertheless, the recent publication of the P. vivax reference genome now allows genomics and systems biology approaches to be applied to this pathogen. We show here that whole genome analysis of the parasite can be achieved directly from ex vivo-isolated parasites, without the need for in vitro propagation. more...

P. vivax: proof-of-principle global transcription analysis from two wild isolates

(Submitter supplied) The complete genome sequence of the P. vivax Sal-1 strain allowed the design of a first version array representing 1 oligo/2 kb of coding sequences (http://zblab.sbs.ntu.edu.sg/vivax/index.html). Here, proof-of-principle experiments using total RNA of parasites obtained from the Sal-1 strain, from P. falciparum and from parasites obtained directly from two human patients are presented.
