Summary |
Using magneto-nanofection, iPS cells were efficiently generated by the transient expression of iPS genes in MEF (mouse embryonic fibroblast), suggesting that the non-viral magneto-nanofection method can be used for the efficient generation of iPS cells.

Overall design |
Plasmid DNAs containing the iPS genes oct4, sox2, klf4 and C-myc were introduced into MEF cells using magneto-nanofection. ESC media were changed daily for two days after transfection, and we could observe some colonies showing ES-like morphology 6-7 days after transfection. The colonies were picked up 12 days after transfection, and then each colony was separately transferred to a new dish containing STO feeder cells.
