Title |
fresh old islets, biological rep1 |
Sample type |
RNA |
Source name |
Fresh old mice islets
Organism |
Mus musculus |
Characteristics |
tissue: pancreatic islets strain: C57BL6 mice age: 56-week-old
Treatment protocol |
Islets were cultured in supplemented RPMI medium for two days at different glucose concentrations (3 mM, 5.5 mM, 11mM, and 16 mM) before RNA extraction
Growth protocol |
Mouse pancreatic islets were isolated from wild-type C57/Bl6J male mice. Islets were allowed to recover for 1 day in RPMI (Invitrogen) containing 11mM glucose supplemented with 10% FCS and penicillin/streptomycin (Invitrogen) at 37°C, 5% CO2.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol through two cycles of cDNA synthesis (50 ng starting total RNA).
Hybridization protocol |
Following fragmentation, cRNA were hybridized, washed and stained according to the standard Affymetrix protocol.
Scan protocol |
GeneChips were scanned according to the standard Affymetrix protocol (Affymetrix Genechip Command Console program 3.0.0.1214, scanner GCS 3000-7G)
Description |
Gene expression data from fresh old islets
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 150.
