Sample GSM1096965 Query DataSets for GSM1096965
Status Public on Mar 25, 2014
Title Paucigranulocytic Asthma 4135447179_B
Sample type RNA

Source name induced sputum
Organism Homo sapiens
Characteristics asthma phenotype: Paucigranulocytic Asthma
gender: Female
smoking pack years: 0
age: 53
sample type: sputum
disease state: asthma
Extracted molecule total RNA
Extraction protocol RNA was extracted from induced sputum using the RNeasy Mini Kit (Qiagen, Hilden, Germany). RNA quatitation was performed using the Quanti-iT RiboGreen RNA Quantitation Assay Kit (Molecular Probes Inc, Invitrogen, Eugene, OR, USA).
Label biotin
Label protocol 500ng extracted RNA was reverse-transcribed into cRNA and labeled with biotin-uridine-5'-triphosphate by using the Illumina TotalPrep RNA Amplication Kit.

Hybridization protocol 750ng reverse-transcribed cRNA was hybridized to the Illumina Sentrix HumanRef-8 Version 2 Expression BeadChip, which measured the expression of 20590 genes.
Scan protocol BeadChips were scanned using the Illumina Bead Station and data was captured using VeadScan 3.5.11.
Description 4135447179_B
Data processing Sample and gene profiling results were included in the analysis if the sample was of suitable purity (OD 260/280 ratio between 1.7-2.1) and was successfully amplified (sufficient cRNA generated) and hybridized (95th percentile of fluorescence score>500) and the data passed quality control criteria in GeneSpring GX (correlation coefficients and principle component analysis plots)
