Title |
2 hours (100 ng/ml FGF2) (2) |
Sample type |
RNA |
Source name |
Lens primary cultures
Organism |
Rattus norvegicus |
Characteristics |
tissue: lens explant age: 3-day old pups
Treatment protocol |
Rat lens explants were grown overnight in the presence of 5 ng/ml FGF2 (reference sample). The cultures were subsequently treated with 100 ng/ml of recombinant FGF2.
Growth protocol |
Rat lens explants were prepared from 3-day old newborn pups. Six explants were used per individual assay.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using RNeasy micro-elute columns (QIAGEN), according to manufacturer's instructions.
Label |
biotin
Label protocol |
50 ng total RNA was amplified using the NUGEN Ovation™ Biotin RNA Amplification and Labeling System. Amplified cDNA was fragmented and biotin labeled according to manufacturer's instructions.
Hybridization protocol |
2.2 micrograms of cDNA was hybridized for 17 hr to the Affymetrix Rat Genome 230 2.0 Arrays as described in the Ovation™ Biotin RNA Amplification and Labeling System manual. Genechips® were processed using protocol EukGE_WS2v4 450 for Genechip® fluidics station FS 450.
Scan protocol |
Affymetrix GeneArray scanner 7G
Description |
2 hours treated lens extracted total RNA_t2h
Data processing |
Feature intensity was extracted by GeneChip Operating System as CEL files. The probe-level analysis of the CEL files was done by the RMA algorithm using GeneSpring GX11 program. No further adjustments were made to the data in the table.
