Treatment protocol |
Freshly thawed parasites were injected into naïve C57BL/6 mice to initiate infection. An inoculum containing 1x10^6 infected red blood cells (iRBCs) suspended in 100 µl phosphate buffer saline (PBS), pH 7.4, from the donor mice was injected intravenously into experimental mice. Naïve mice receiving an equivalent number of uninfected RBCs served as a negative control group.

Inbred female C57BL/6 mice, aged 6-8 weeks old, were purchased from Charles River Laboratories International, Inc. (Frederick, Maryland, USA), Jackson Laboratory (Bar Harbor, Maine, USA), or were obtained from the NIAID/Taconic repository.

Four days after parasite injection, spleens from infected or uninfected mice were removed and preserved in RNA Later solution (Qiagen, Valencia, California, USA). RNA was isolated using an RNeasy mini kit following the manufacturer's protocol (Qiagen).
