Treatment protocol A 50 mM stock solution of Berberine chloride (Sigma-Aldrich) was prepared in DMSO. Cells were treated with 40 M berberine chloride or 0.08% DMSO as control. Comparing miRNA profiles of 40 M berberine-treated PHHs to those of control cells sampled after 2, 4, and 8 hours treatment
Growth protocol The primary human hepatocytes (induction qualified human plateable cryopreserved hepatocytes, HMCPIS) from Caucasian donors were purchased from Invitrogen and cultured following the manufacturer’s instructions.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using TRIZOL reagent (Invitrogen) according to the manufacture’s protocol. The total RNA quantity was measured using a NanoDrop ND-1000 Spectrophotometer (Nanodrop Technologies). The total RNA quality and integrity were assayed by Agilent 2100 bioanalyzer with RNA 6000 nano kit (Agilent Technologies).

Data processing The data were normalised using quantile normalization with by GeneSpring GX.
