cKO females were injected intraperitoneally with vehicle (saline) or E2 (0.25 ug of 17beta-estradiol) for 2 h or 24 h.

Growth protocol |
cKO females at the age of 8 to 10 weeks were ovariectomized and housed for 10 days before the treatment.

Extraction protocol |
Uterine samples were collected at 2 or 24 h after the injection. Tissues were immediately frozen in liquid nitrogen. RNA was extracted by using RNeasy Mini Kit (Qiagen, Valencia, CA) and treated with DNaseI (Qiagen) according to the manufacturer's protocol.

Starting with 500ng of total RNA, Cy3 labeled cRNA was produced according to manufacturer's protocol.

For each sample, 1.65ug of Cy3 labeled cRNAs were fragmented and hybridized for 17 hours in a rotating hybridization oven.

Slides were washed and then scanned with an Agilent Scanner.
