Title |
B6.Lb_1 |
Sample type |
RNA |
Source name |
whole mouse ear with Leishmania lesion
Organism |
Mus musculus |
Characteristics |
strain of mouse: C57BL/6 species of leishmania: braziliensis susceptibility phenotype: resistant tissue: whole mouse ear
Treatment protocol |
Ear tissue was preserved in RNAlater reagent (Qiagen) and stored at -80c.
Growth protocol |
6-8wk old mice were infected with 105 metacyclic-enriched parasites. Mice were killed at 4 weeks post-infection and the infected ear was recovered.
Extracted molecule |
total RNA |
Extraction protocol |
RNAlater-preserved ear tissue was weighed, minced with a razor blade, and the entire ear was homogenized in 1ml of Qiazol (Qiagen) using 3 x 20 second pulses on a FastPrep-24 MP homogenizer (MP Biomedicals), with 5 min on ice between each pulse. Total RNA was isolated using the Qiagen miRNeasy kit.
Label |
biotin
Label protocol |
Biotin labeled complementary RNA (cRNA) was made using the EpiCentre Nano Total-Amp kit
Hybridization protocol |
Standard Illumina hybridization protocol
Scan protocol |
Standard Illumina scanning protocol
