Worms were grown on RNAi plates from egg stage until fifth day of adulthood. Wild type N2 and isp-1(qm150);ctb-1(qm189) were grown on control L4440 plates as control. isp-1(qm150);ctb-1(qm189) was also grown on klf-1 RNAi plates until larval L4 stage and then switched to L4440 plates, or switched at L4 larval stage to klf-1 RNAi plates from control L4440 plates.

RNA was isolated using Rneasy Mini Kit (QIAGEN) according to manufacturors protocol. The samples were amplified with the NuGen Applause WT-Amp Plus ST Kit.
